摘要
由青枯雷尔氏菌(Ralstonia solanacearum)引发的青枯病,是马铃薯第一大细菌性病害.通过生理生化与时空转录组分析,揭示抗病种质C804抵御青枯菌P2侵染的多层次协同防御机制.结果表明C804通过“酶-代谢-基因”三位一体网络实现高效抗性.生理层面,其超氧化物歧化酶和过氧化物酶的级联反应精准清除活性氧,结合脯氨酸代谢重编程维持氧化稳态;而易感种质A056因酶活性滞后及膜脂过氧化加剧导致防御失调.分子层面,C804通过ScPR1-ScMLP1双基因模块构建时空防御:与A056相比,ScPR1在其根和茎组织分别呈现6.2倍和8.5倍爆发式表达,激活系统获得性抗性;ScMLP1在侵染中期特异性高表达(根部78.9倍,茎部239.3倍),强化物理屏障.
Bacterial wilt caused by Ralstonia solanacearum is the most devastating bacterial disease affecting potato(Solanum tuberosum L.).This study unraveled the multi-layered synergistic defense mechanisms of resistant germplasm C804 against R.solanacearum strain P2 through physiological,biochemical,and spatiotemporal transcriptomic analyses.Results demonstrated that C804 achieves high-efficiency resistance via an enzyme-metabolism-gene three-in-one network.At the physiological level,its SOD and POD cascade precisely scavenges ROS,while proline metabolic reprogramming maintains oxidative homeostasis.Susceptible genotype A056 exhibits delayed enzymatic activity and aggravated membrane lipid peroxidation,leading to defense dysregulation.At the molecular level,C804 establishes spatiotemporal defense through the ScPR1-ScMLP1 dual-gene module:ScPR1 shows explosive upregulation in roots(6.2-fold)and stems(8.5-fold),activating systemic acquired resistance,while ScMLP1 specifically overexpresses at mid-infection stage(78.9-fold in roots,239.3-fold in stems)to reinforce physical barriers.
