摘要
目的采用低共熔溶剂(DES)超声辅助技术提取山银花中绿原酸(CGA),建立最佳DES提取方案,并初步明确DES回收的山银花提取物的体外抗氧化活性。方法通过高效液相色谱(HPLC)法测定提取物中CGA的含量,依据含量高低对比从30种DES中筛选出提取山银花中CGA效果最佳的DES。研究摩尔比、含水量、液固比、提取时间、超声功率、提取温度对山银花CGA提取效果的影响,进一步运用响应面法优化提取条件,并与超声法(UAE)、浸提法(ME)、渗漉法(PE)进行对比。最后以维生素C和槲皮素为对照,研究DES回收的山银花CGA提取物的体外抗氧化活性。结果对不同提取条件得到的CGA类化合物的含量进行比较,得到最佳的DES为L-脯氨酸和L-乳酸混合物,最佳提取条件为:DES为L-脯氨酸∶L-乳酸=1∶3,含水量为30%(体积分数),液固比为16 mL/g,提取时间为25 min,提取温度为45℃,超声功率为240 W;CGA最大提取率为4.53%±0.11%。与传统提取方法相比,DES提取效果最佳。DES回收的山银花CGA提取物具有一定的体外抗氧化活性,其清除率约为同等浓度下的维生素C或槲皮素清除率的40%~50%。结论DES是一种绿色、高效提取山银花中CGA的溶剂,可为中药活性成分的提取研究提供一定的参考。
Objective To extract chlorogenic acid(CGA)from Lonicerae Flos using deep eutectic solvents(DES)assisted by ultrasound technology,and to establish the optimal DES extraction scheme and preliminarily clarify the in vitro antioxidant activity of Lonicerae Flos extract recovered by DES.Methods The content of CGA in the extract was determined by high-performance liquid chromatography(HPLC).The DES with the best CGA extraction effect was selected from 30 DES according to the content comparison.The effects of molar ratio,water content,liquid-solid ratio,extraction time,ultrasonic power and extraction temperature on CGA extraction were studied.Response surface methodology was further used to optimize the extraction conditions,and the results were compared with ultrasonic method(UAE),maceration method(ME)and percolation method(PE).Finally,the in vitro antioxidant activity of the Lonicerae Flos extract recovered by DES was investigated using vitamin C and quercetin as controls.Results Comparison of the contents of CGA compounds obtained under different extraction conditions showed that the best DES were L-proline and L-lactic acid mixture,with the best extraction conditions being L-proline∶L-lactic=1∶3,30%water content,a liquid-solid ratio of 16 mL/g,an extraction time of 25 min,an extraction temperature of 45℃,and an ultrasonic power of 240 W.The maximum CGA extraction rate was 4.53%±0.11%.Compared with traditional extraction methods,the DES extraction was the most effective.The comparison with vitamin C and quercetin indicated that the Lonicerae Flos extract recovered by DES had certain in vitro antioxidant activity,and its clearance was approximately 40%~50%of that of vitamin C or quercetin at the same concentration.Conclusions DES is a green and efficient solvent for extracting CGA from Lonicerae Flos,which can provide a reference for the extraction of active components of traditional Chinese medicine.
