花椒锈病抗性相关的SNP分子标记筛选

摘要 【目的】开发和筛选与花椒锈病抗性相关的SNP分子标记,实现不同花椒品种(系)锈病高效精准的抗性分子鉴定。【方法】基于花椒转录组测序(RNA-Seq)结果,开发出可能与抗性相关的SNP标记,在抗感差异材料中验证与花椒锈病抗性相关的SNP引物。【结果】以12份抗病株系和感病株系基因组DNA为模板,与人工接种鉴定相结合,获得1个与花椒抗锈病相关的SNP标记,该标记在抗病材料中能够扩增出1条325 bp左右大小的条带(命名为30616-314-2)。使用此引物对现有40份花椒种质资源进行抗性鉴定,获得抗病材料14份,感病材料26份。【结论】筛选出了1个可以用于花椒抗锈病辅助育种选择的SNP分子标记,加速推进花椒抗锈病育种进程。 【Objective】To develop and screen SNP molecular markers associated with rust disease resistance to in Zanthoxylum bungeanum,and to facilitate efficient and accurate molecular identification of rust resistance in Z.bungeanum varieties(lines).【Method】Based on RNA-Seq data,SNP markers potentially linked to rust resistance were developed.Subsequently,the primers for these SNP markers were validated in both resistant and susceptible Z.bungeanum materials.【Result】A SNP marker associated with rust resistance in Z.bungeanum was identified from the genomic DNA of 12 lines,which included both resistant and susceptible individuals.Using this marker in conjunction with artificial inoculation tests,a 325 bp DNA band(designated as 30616-314-2)was amplified specifically in resistant materials.A total of 40 Z.bungeanum germplasm resources were screened for rust resistance using this primer,resulting in the identification of 14 resistant and 26 susceptible materials.【Conclusion】A SNP marker has been successfully developed for the molecular assistance in breeding rust-resistant Z.bungeanum,which is expected to accelerate the breeding process for rust resistance in this species.
