摘要
为研究太子参皂苷(pseudostellaria heterophylla saponins,PHS)对模式抗原卵清白蛋白(OVA)免疫小鼠肠道免疫应答的影响,将72只雌性小鼠随机分为6组,分别为空白组(CK组)、OVA免疫对照组(OVA组)、铝佐剂对照组(OVA+Al组)、PHS低剂量组(OVA+PHS-L,50 mg/kg)、PHS中剂量组(OVA+PHS-M,100 mg/kg)、PHS高剂量组(OVA+PHS-H,200 mg/kg)(在第1~5天和第16~20天,除PHS各剂量组按体重灌胃相应剂量PHS外,其余各组灌胃相应体积的双蒸水;在第6天和第21天,于腹股沟处对CK组每只小鼠进行皮下注射0.2 mL/只的无菌PBS,对OVA+Al组注射含100μg OVA和200μg Alum的PBS混合液,对PHS各剂量组小鼠注射含100μg OVA的PBS溶液)。第36天处死小鼠,检测各项指标。结果显示,与OVA组相比,OVA+PHS-L组可显著提高回肠绒隐比和十二指肠单位长度内淋巴细胞个数(P<0.05),显著提升十二指肠中IL-6、空肠中sIgA和回肠中TNF-α含量(P<0.05);OVA+PHS-M组可显著提高十二指肠绒毛高度、绒隐比以及单位长度内淋巴细胞个数(P<0.05),显著提升空肠和回肠中TNF-α含量(P<0.05);OVA+PHS-H组可显著提升空肠中sIgA含量(P<0.05)。上述结果表明,PHS能在一定程度上强化肠道组织形态,调节相关免疫球蛋白和细胞因子的分泌,增强肠道黏膜免疫功能,提高小鼠对OVA的免疫应答,具有成为佐剂的潜力。
The aim of this study was to investigate the effects of Pseudostellaria heterophylla saponins(PHS)on intestinal immune response in mice immunized with model antigen ovalbumin(OVA).Seventy-two female mice were randomly divided into 6 groups:blank group(CK group),OVA immune control group(OVA group),aluminum adjuvant control group(OVA+Al group),PHS low-dose group(OVA+PHS-L,50 mg/kg),PHS medium-dose group(OVA+PHS-M,100 mg/kg),PHS high-dose group(OVA+PHS-H,200 mg/kg).On the 1st to 5th day and the 16th to 20th day,the PHS groups were intragastrically administered with the corresponding dose of PHS according to body weight,the other groups were intragastrically administered with the corresponding volume of double distilled water;on the 6th and 21 st day,the mice in the CK group were subcutaneously injected with sterile PBS at a dose of 0.2 mL/mouse at the groin,the OVA+Al group was injected with PBS mixture containing 100μg OVA and 200μg Alum,and the PHS groups were injected with PBS solution containing 100μg OVA.On the 36 th day,the mice were sacrificed and the indexes were detected.The results showed that compared with the OVA group,the OVA+PHS-L group significantly increased the the ratio of ileum villus to crypt and the number of lymphocytes per unit length of duodenum(P<0.05),and significantly increased the content of IL-6 in duodenum,sIgA in jejunum and TNF-αin ileum(P<0.05);the OVA+PHS-M group could significantly increase the height of duodenal villus,the ratio of villus to crypt and the number of lymphocytes per unit length(P<0.05),and significantly increase the content of TNF-αin jejunum and ileum(P<0.05);OVA+PHS-H group could significantly increase the content of sIgA in jejunum(P<0.05).The above results show that PHS can strengthen the intestinal tissue morphology to a certain extent,regulate the secretion of related immunoglobulins and cytokines,strengthen the intestinal mucosal immune function,improve the immune response of mice to OVA,and has the potential to become an adjuvant.
