姜黄素-IR1061新型纳米脂质体的制备及其对肝癌细胞的体外抗肿瘤/光热联合治疗研究

摘要 目的制备一种共同负载姜黄素和IR1061染料的新型纳米脂质体,并研究其对肝癌细胞的抗肿瘤/光热联合治疗效果。方法采用薄膜水化法制备脂质体IR1061@姜黄素-Lips。对其粒径大小、稳定性、表面Zeta电位、多分散系数(PDI)、包封率、光热转换能力及光热稳定性进行考察;细胞水平上,利用激光扫描共聚焦显微镜探讨肝癌细胞对IR1061@姜黄素-Lips的摄取行为;通过细胞毒性实验,评估IR1061@姜黄素-Lips对肝癌细胞的抗肿瘤+光热联合治疗效果。结果IR1061@姜黄素-Lips的平均粒径(176.77±10.17)nm,分布均匀,稳定性良好,姜黄素药物的包封率为(64.79±2.62)%;带负电位,Zeta电位为(-37.17±0.41)mV;PDI为(0.242±0.018);在近红外808 nm激光照射下具有良好的光热升温效果,光热稳定性好,计算得光热转换效率高达47.45%;细胞实验结果表明脂质体的形式增加了肝癌细胞对药物的摄取,姜黄素与IR1061的联合对肝癌细胞有显著的抗肿瘤/光热协同抑制作用。结论成功制备了新型纳米脂质体IR1061@姜黄素-Lips,证明了其对肝癌细胞有显著的抗肿瘤/光热协同抑制作用,为肝癌患者在治疗方式的选择上提供了一种新的可能。 Objective To prepare a novel nanoliposome loaded with curcumin and IR1061 dye,and to study its anti-tumor/photothermal combined therapy effect on hepatocellular carcinoma cells.Methods Liposomes IR1061@curcumin-Lips were prepared by thin film hydration.The particle size,stability,surface Zeta potential,polydispersity index(PDI),encapsulation efficiency,photothermal conversion ability,and photothermal stability were investigated.At the cellular level,laser scanning confocal microscopy was used to investigate the uptake behavior of IR1061@curcumin-Lips by hepatocellular carcinoma cells.The effect of IR1061@curcumin-Lips combined with anti-tumor and photothermal on hepatocellular carcinoma cells was evaluated by cytotoxicity test.Results The average particle size of IR1061@curcumin-Lips was(176.77±10.17)nm,which was uniform distribution and good stability.The encapsulation efficiency of curcumin was(64.79±2.62)%.The Zeta potential was(-37.17±0.41)mV.PDI was(0.242±0.018);it had a good photothermal heating effect and good photothermal stability under the near-infrared 808 nm laser irradiation,and the photothermal conversion efficiency was as high as 47.45%.The results of cell experiments showed that the liposome form of curcumin increased the drug uptake of hepatoma cells,and the combination of curcumin and IR1061 had a significant anti-tumor/photothermal syner-gistic inhibition effect on hepatocellular carcinoma cells.Conclution A novel nano-liposome IR1061@curcumin-Lips has been successfully prepared,and it has a significant anti-tumor/photothermal synergistic inhibition effect on liver cancer cells,which is expected to provide a new idea for the treatment of liver cancer.
