黄芪甲苷、β-榄香烯增强小鼠巨噬细胞免疫功能的体外实验研究 被引量:20

摘要 目的观察黄芪甲苷、β-榄香烯对小鼠巨噬细胞(Mφ)免疫功能的影响,为阐明黄芪、莪术的抗肿瘤机制提供实验依据。方法黄芪甲苷、β-榄香烯体外作用小鼠Mφ细胞株J447A.1细胞,用流式细胞仪检测其表面分子(MHCⅡ、MHCⅠ、CD40、CD86),吞噬中性红实验检测Mφ的吞噬能力。结果经黄芪甲苷、β-榄香烯处理,J447A.1细胞表面MHCⅡ、MHCⅠ、CD40、CD86分子的表达水平均高于空白组。其中黄芪甲苷组MHCⅠ水平明显高于空白组(P<0.05);β-榄香烯组CD40表达明显高于空白组(P<0.05);黄芪甲苷联合β-榄香烯组MHCⅡ、CD86表达明显高于空白组(P<0.05)。黄芪甲苷作用12h后,Mφ吞噬中性红的能力明显高于正常对照组(P<0.05);β-榄香烯作用24h后,Mφ吞噬中性红的能力明显高于正常对照组(P<0.05),尤以100μg/ml的作用浓度最佳;经黄芪甲苷与β-榄香烯联合作用后,Mφ吞噬中性红的能力明显高于正常对照组(P<0.05)。结论黄芪甲苷、β-榄香烯及其联用能增强Mφ表面MHC分子和免疫共刺激因子表达,促进Mφ的吞噬能力。 Objective To observe the influence of Astragaloside IV and β-elemene on immunological function of macrophage,and explore the experimental basis of anti-tumor mechanism of Astragaloside IV and β-elemene.Methods J447A.1 cells,the mouse macrophage strain,was exposed to Astragaloside IV and β-elemene in vitro.The cell surface molecules,MHCⅡ,MHCⅠ,CD40,CD86,were detected by flow cytometry(FCM).The phagocytic capability of macrophage was determined by neutral red dye test.Results Following the treatment of Astragaloside IV and β-elemene in vitro,the levels of MHCⅡ,MHCⅠ,CD40,CD86 on J447A.1 cell surface were significantly higher than those of the control group.The level of MHCⅠ in Astragaloside IV group was obviously higher than that in control group(P0.05).The level of CD40 in β-elemene group was obviously higher than that in control group(P0.05).The levels of MHCⅡ and CD86 in both of Astragaloside IV and β-elemene group were obviously higher than those in control group(P0.05).In 12 h after treatment by Astragaloside IV,the phagocytic ability of macrophages for neutral red dye was obviously higher than that in control group(P0.05).In 24 h after treatment by β-elemene,the phagocytic ability of the macrophages for neutral red dye was obviously higher than that in control group(P0.05).The concentration of 100 μg/ml of Astragaloside IV or β-elemene was the optimum for the treatment.After treatment of both Astragaloside IV and β-elemene the phagocytic function of macrophages for neutral red dye was obviously higher than that in control group(P0.05) with the increase of drug concentration and reactive time.Conclusion Astragaloside IV and β-elemene as well as the combination of the two drugs could enhance the level of MHC molecules and co-stimulatory factors on the surface of macrophages and elevate the phagocytic ability of macrophages.
