摘要
【目的】利用香蕉细菌性软腐病菌XJ8-3-3全基因组测序结果对其信号肽进行预测和分析,为深入研究其分泌蛋白、明确香蕉与病原微生物互作的分子机制奠定基础。【方法】应用Signal Pv4.1、TMHMMv2.0、THUMBUP、Target P1.1、Lipo P1.0、Tat P1.0、big-PI predictor 7个软件对香蕉细菌性软腐病菌基因组中4 313个ORFs进行预测分析,并通过swisspot数据库预测信号肽所在ORFs的功能。【结果】香蕉细菌性软腐病菌中存在具有信号肽的ORFs 218个,占其基因组编码ORFs的5.05%,其中有184个ORFs属于Ⅰ型信号肽,32个ORFs属于Ⅱ型信号肽,2个ORF为CYT型,未发现Prepilin-like型信号肽的存在。在所有分泌蛋白中,有122个具有可预测的功能,其余96个为功能未知的推定蛋白。【结论】解析了香蕉细菌性软腐病菌基因组ORFs中的信号肽及分泌蛋白的可能功能,发现存在很多果胶酶、蛋白酶、纤维素酶等致病因子和一系列其他可能的致病因子,这为后续致病机理的研究提供了众多候选基因。
【Objective】According to the whole-genome sequencing results of soft rot bacteria XJ8-3-3 inbanana,Prediction and analysis of signal peptide for this bacteria can help us to research secreted pro-teins,which benefits confirming the molecular mechanisms of the interaction between banana and patho-genic microorganisms.【Method】Using several software of Signal Pv4.1,TMHMMv2.0,THUMBUP,Tar-get P1.1,Lipo P1.0,Tat P1.0 and big-PI predictor,4 313 ORFs of the genome of banana soft rot bacteriawere predicted and analyzed. In addition,the function of ORFs where were the signal peptides was pre-dicted by Swissprot database.【Result】We found that there were 218 ORFs containing signal peptide ac-counting for 5.05% of the ORFs coded by genome,in which there were 184 ORFs belonging to typeⅠ,32 ORFs belonging to type Ⅱ,2 ORFs belonging to type CYT,but Prepilin-like type was not discov-ered. Signal peptides had 22.56 amino acids evenly,and most of all were nonpolar. In all secreted pro-teins,122 had divinable function,and others were function-unknown putative protein.【Conclusion】Theanalysis of the signal peptide and secreted proteins in banana soft rot bacteria XJ8-3-3 genome ORF re-vealed a lot of pathogenic factors such as pectinase,protease,cellase and some assumed pathogenic fac-tors wich could act as candidate genes for the further studies of the interaction between banana and patho-genic bacteria: Banana soft rot bacteriaSignal peptideSecreted protein
