PA@Ga-CMCs/GL多孔敷料的制备与抗菌性能研究

摘要 细菌感染是慢性伤口形成的主要原因之一,如果不及时处理会引发伤口持续性炎症和组织受损,严重者面临截肢甚至是死亡的风险。然而,过度和长期使用抗生素等抗菌药物容易造成耐药性细菌滋生。因此,迫切需要开发一种持久高抗菌活性的新型伤口敷料。以生物可吸收的羧甲基壳聚糖(CMCs)和明胶(GL)为基本原料,采用原儿茶醛和金属离子Ga^(3+)三元络合物(PA@Ga)为交联剂构筑了凝胶交联网络,再通过冷冻干燥的手段制备了具有持久高抗菌活性的多孔伤口敷料PA@Ga-CMCs/GL。通过扫描电镜证明了抗菌敷料具有连通多孔的结构,定性和定量抗菌实验表明抗菌敷料对大肠杆菌和枯草芽孢杆菌的抑菌率均达到了96%以上。将PA@Ga-CMCs/GL浸泡去离子水中24 h后,再次冻干得到的多孔材料仍具有高抗菌性能。此外,PA@Ga-CMCs/GL浸泡24 h后的浸提液也表现出一定的抗菌活性。工作中高抗菌活性多孔敷料为细菌感染型伤口的愈合提供了有效的解决方法。 Bacterial infections are one of the main causes of chronic wounds formulation.It causes persistent inflammation and tissue damage around the wound,even lead to amputation risks or even death.However,the excessive and prolonged use of antibiotics and other antibacterial drugs can easily lead to the growth of drugresistant bacterial strains.Therefore,there is an urgent requirement to develop a novel wound dressing with persistent high antibacterial activity.In this paper,with bioabsorbable carboxymethyl chitosans(CMCs)and gelatin(GL)as the basic raw materials,protocatechualdehyde and metal ion Ga^(3+)ternary complex(PA@Ga)as crosslinking agents agel crosslinking network was constructed.Porous wound dressings,PA@Ga-CMCs/GL,was obtained by freezedrying the gel crosslinking network.Scanning electron microscopy characterization demonstrated that the antibacterial dressing possesses a connected and porous structure.Qualitative and quantitative antibacterial experiments showed that the antimicrobial dressing reach a bacteriostasis rate of more than 96%against Escherichia coli and Bacillus subtilis.After been immersed into deionized water for 24 h,the PA@Ga-CMCs/GL still showed high antibacterial properties.The water extract also showed an antibacterial performance.The highly antibacterial active dressing provides a promising protentional for bacterial infected wound healing.
