光唇鱼fads2基因克隆及其在不同温度对比下的表达

摘要 为探究光唇鱼不饱和脂肪酸合成关键酶脂肪酸去饱和酶2(fads2)对鱼类不饱和脂肪酸合成的调控,首先对fads2基因克隆,得到克隆基因序列后对fads2基因的结构进行了生物信息学分析,并且在不同温度梯度下饲养光唇鱼,使用实时荧光定量(qRT-PCR)测定不同温度下fads2基因的表达量差异,对比不同温度下fads2基因的表达量。生物信息学分析该基因为亲水性蛋白,结构稳定,含有与脂肪酸合成有关的Cyt-b5结构域。从fads2的定量表达结果来看,在28℃条件下fads2表达量显著上调(P<0.05),研究表明,fads2基因的表达量与温度有关。研究结果有助于深入了解光唇鱼fads2基因的结构和功能,为进一步的研究鱼类脂质代谢提供理论基础,从而选育出高不饱和脂肪酸合成较高的优良光唇鱼品种。 In order to explore the regulation of unsaturated fatty acid synthesis by fatty acid desaturase 2(fads2),a key enzyme of unsaturated fatty acid synthesis in Acrossocheilus fasciatus,Firstly,fads2 gene was cloned,the cloned gene sequence was obtained,and the structure of the fads2 gene was analyzed bioinformatics.Besides,A.fasciatus were fed under different temperature gradients.Real-time fluorescence quantification(qRT-PCR)was used to determine the difference of fads2 gene expression at different temperatures,and compared the fads2 gene expression at different temperatures.Bioinformatics analysis showed that the gene was a hydrophilic protein with stable structure and a Cyt-b5 domain related to fatty acid synthesis.According to the quantitative expression results of fads2,the expression level of fads2 was significantly up-regulated at 28℃(P<0.05),and the study showed that the expression level of fads2 gene was related to temperature.The results of this experiment are helpful to further understand the structure and function of fads2 gene in A.fasciatus,and provide a theoretical basis for further research on lipid metabolism of fish,so as to select excellent Acrossocheilus species with high synthesis of unsaturated fatty acids.
