摘要
利用酶-超声辅助法提取秭归脐橙皮中的黄酮类化合物,具有操作方便、节能高效的优点。以秭归脐橙皮为原料利用酶辅助超声提取法通过单因素试验和正交试验研究其提取方法,同时对黄酮降血糖的作用进行研究。结果表明,最佳提取工艺为酶添加量(与原料质量之比)为0.50%,乙醇体积分数为50%,料液比为1∶15(g∶mL),超声时间为10 min。在影响提取率的因素中,料液比>乙醇体积分数>超声时间>酶添加量。降血糖研究表明,当黄酮供试液质量浓度为4 mg/mL时,对α-葡萄糖苷酶溶液抑制率可达到50%以上;当黄酮供试液质量浓度为10 mg/mL时抑制率可达到68.67%。当黄酮供试液质量浓度为4 mg/mL时,对α-淀粉酶溶液的抑制率可达到50%以上;当黄酮供试液质量浓度为10 mg/mL时抑制率可达到74.81%。由此可知,秭归脐橙皮中的黄酮类化合物具有良好的降血糖作用。
Extracting flavonoids from Zigui navel orange peel by enzyme-ultrasonic assisted method has the advantages of convenient operation and energy saving.In this paper,the enzymatic assisted ultrasonic extraction method of Zigui navel orange peel was used as raw material to study its efficient extraction method through four single factor and orthogonal test,and the effect of flavonoids on lowering blood sugar was also studied.The results showed that the optimal extraction process was 0.50%enzyme dosage(ratio of raw material to mass),50%ethanol concentration,1∶15(g∶mL)solid-liquid ratio,and 10 min ultrasonic time.Among the factors affecting the extraction rate,the liquid-solid ratio>ethanol concentration>ultrasonic time>enzyme dosage.The hypoglycemic study showed that the inhibition rate ofα-glucosidase solution could reach more than 50%when the concentration of flavonoid solution was 4 mg/mL,and the inhibition rate ofα-glucosidase solution could reach 68.67%when the concentration of flavonoid solution was 10 mg/mL.When the concentration of flavonoid was 4 mg/mL,the inhibition rate ofα-amylase solution could reach more than 50%,and when the concentration of flavonoid solution was 10 mg/mL,the inhibition rate ofα-amylase solution could reach 74.81%.It could be seen that flavonoids in Zigui navel orange peel had good hypoglycemic effect.
