祛风湿中药威灵仙、豨莶草对痹证大鼠作用的性-效关系研究 被引量:5

Property-efficacy relationship of antirheumatic traditional chinese medicine Clematidis Radix et Rhizoma and Siegesbeckiae Herba on arthralgia syndrome rats

摘要 目的:通过对比祛风湿温性中药威灵仙、寒性中药豨莶草对风寒湿痹、风湿热痹类风湿关节炎(rheumatoid arthritis,RA)模型大鼠的作用差异,探讨中药药性-病证-疗效之间的关联性。方法:将112只SD大鼠按随机数字表法分为空白组、Ⅱ型胶原(CⅡ)组、风寒湿模型(FHS-M)组、风寒湿中药干预(包括FHS-L、FHS-WG、FHS-WD、FHS-XG、FHS-XD)组、风湿热模型(FSR-M)组、风湿热中药干预(包括FSR-L、FSR-WG、FSR-WD、FSR-XG、FSR-XD)组,每组8只。除空白组外,其余采用天然牛Ⅱ型胶原蛋白及施加风寒湿、风湿热环境因素刺激诱导造模。实验中记录各组大鼠一般状态、关节肿胀度和关节炎评分,HE染色法观察大鼠踝关节组织病理形态变化,ELISA法检测大鼠血清TNF-α、ICAM-1、IL-6含量,IHC法检测大鼠踝关节VEGF-A、MMP-3、TIMP-1蛋白表达,RT-PCR法检测大鼠踝关节VEGF-A、MMP-3、TIMP-1 mRNA表达。结果:与空白组比较,CⅡ组、FHS-M组、FSR-M组大鼠足跖肿胀度、AI评分显著升高,踝关节组织结构明显破坏,血清TNF-α、ICAM-1含量显著升高,VEGF-A、MMP-3 mRNA水平及蛋白表达水平明显升高,TIMP-1 mRNA水平及蛋白表达水平明显降低。与FHS-M组或FSR-M组比较,各给药组大鼠足跖肿胀度、AI评分显著降低,踝关节组织病理结构明显减轻,血清TNF-α、ICAM-1含量显著降低,VEGF-A、MMP-3 mRNA水平及蛋白表达水平明显降低,TIMP-1 mRNA水平及蛋白表达水平明显升高,其中与FHS-M组比较,FHS-XG组对RA大鼠的改善幅度最大;与FSR-M组比较,FSR-XG组对RA大鼠的改善幅度最大。结论:威灵仙、豨莶草均能改善RA大鼠的病情,但威灵仙对风寒湿痹RA大鼠的治疗效果比豨莶草更好,豨莶草对风湿热痹RA大鼠的治疗效果比威灵仙更好。 OBJECTIVE To compare the differences in the effects of Clematidis Radix et Rhizoma and Siegesbeckiae Herba on rheumatoid arthritis(RA)model rats with wind-cold-dampness/wind-dampness-heat arthralgia and examine the correlation with the nature of traditional Chinese medicine-disease-syndrome-efficacy.METHODS A total of 112 Sprague-Dawley(SD)rats were randomized into 14 groups of blank,type Ⅱ collagen(CⅡ),wind-cold-dampness model(FHS-M),wind-colddampness Chinese medicine intervention(including FHS-L,FHS-WG,FHS-WD,FHS-XG&FHS-XD),wind-dampnessheat model(FSR-M),wind-dampness-heat Chinese medicine intervention(including FSR-L,FSR-WG,FSR-WD,FSR-XG&FSR-XD)(n=8 each).In addition to blank group,the remainders were induced by natural bovine type Ⅱ collagen and a stimulation of wind-cold-dampness and wind-dampness-heat environmental factors.General profiles,joint swelling degree and arthritis score of rats in each group were recorded.The pathological changes of ankle joint tissue were observed by hematoxylin-eosin(HE)stain.The serum contents of TNF-α,ICAM-1 and IL-6 were detected by enzyme-linked immunosorbent assay(ELISA).The expressions of VEGF-A,MMP-3 and TIMP-1 protein in ankle joint were quantified by immunohistochemistry(IHC).The expression of VEGF-A,MMP-3 and TIMP-1 mRNA in ankle joint was detected by real-time polymerase chain reaction(RTPCR).RESULTS As compared with blank group,paw swelling degree and AI score of rats spiked sharply in CⅡ,FHS-M and FSR-M groups.Structural destruction of ankle joint was evident.Serum contents of TNF-α and ICAM-1 rose,mRNA/protein expressions of VEGF-A and MMP-3 spiked and mRNA/protein expression of TIMP-1 dropped.As compared with FHS-M/FSR-M group,paw swelling degree and AI score of rats dropped obviously in each dosing group.Pathological structure of ankle joint tissue diminished,serum contents of TNF-α and ICAM-1 declined,mRNA/protein expressions of VEGF-A and MMP-3 dropped and mRNA/protein expression of TIMP-1 rose.As compared with FHS-M group,FHS-XG group had the greatest improvement.As compared with FSR-M group,FSR-XG group had the greatest improvement.CONCLUSION Both Clematidis Radix et Rhizoma and Siegesbeckiae Herba may improve the status of RA rats.However,the therapeutic effect of Clematidis Radix et Rhizoma is superior to that of Siegesbeckiae Herba.And the therapeutic effect of Siegesbeckiae Herba with winddampness-heat arthralgia is better than that of Clematidis Radix et Rhizoma in RA rats.
