摘要
目的:以旋覆代赭汤乙酸乙酯部位中的多酚类化合物为研究对象,探索中药组方配伍对其含量变化的影响,以及与抗氧化活性的相关性。方法:建立旋覆代赭汤乙酸乙酯部位12种多酚类成分的含量测定方法,采用Waters ACQUITY UPLC BEH C18(2.1 mm×150 mm,1.7μm)色谱柱,流动相为乙腈-甲醇-0.4%甲酸水溶液,梯度洗脱,体积流量0.1 mL/min,柱温40℃,提取二维图谱波长350 nm,随后对旋覆代赭汤不同配伍组进行含量测定。同时检测了不同配伍组的总抗氧化能力(T-AOC),DPPH自由基清除能力以及ABTS自由基清除能力,采用灰色关联度法和偏最小二乘回归分析研究了不同配伍下12种多酚类成分与抗氧化活性的相关性。结果:与单味药组相比,不同配伍组各成分含量变化并不相同。旋覆代赭汤乙酸乙酯部位具有较强的抗氧化活性,且君药旋覆花对抗氧化活性有主要贡献,12种多酚类成分与抗氧化活性关联度均>0.6,绿原酸和异绿原酸B可能是发挥抗氧化活性的关键成分,且配伍对这两种成分的含量均有显著影响。结论:旋覆代赭汤中的多酚类成分与抗氧化活性之间呈线性相关,且多成分共同发挥药效,该研究可为进一步探索旋覆代赭汤药效物质基础研究提供实验依据。
Objective:The study focuses on polyphenols in the ethyl acetate part of Xuanfu Daizhe Decoction(旋覆代猪汤)to explore the influence of the compatibility of traditional Chinese medicine prescriptions on its content change and its correlation with antioxidant activity.Methods:A method for determining the content of 12 kinds of polyphenols in the ethyl acetate part of Xuanfu Daizhe Decoction was established.Waters ACQUITY UPLC BEH C18(2.1 mmx150 mm,1.7μm)column was used,the mobile phase was acetonitril-methanol-0.4%formic acid aqueous solution,and other conditions were gradient elution,volume flow rate of o.1 mL/min,column temperature of 40 C,and 2D spectrum wavelength of 350 nm.Then,the contents of Xuanfu Daizhe Decoction in different compatibility groups were determined.The total antioxidant activity(T-AOC),DPPH free radical scavenging capacity,and ABTS free radical scavenging capacity of different compatibility groups were measured.The correlation between 12 polyphenols and antioxidant activity was studied by the grey correlation method and partial least square regression analysis.Results:Compared with the single drug group,the content changes of each component in dfferent compatibility groups were different.The ethyl acetate part of Xuanfu Daizhe Decoction had strong antioxidant activity,and monarch drug,namely INULAE FLOS had the main contribution to the antioxidant activity.The correlation degree between 12 polyphenols and antioxidant activity was larger than O.6.Chlorogenic acid and isochlorogenic acid B may be the key components to exert antioxidant activity,and their compatibility had a significant influence on the content of these two components.Conclusion:There is a linear correlation between polyphenols and antioxidant activity in Xuanfu Daizhe Decoction,and many components exert their efficacy together.This study can provide an experimental basis for further exploring the material basis of Xuanfu Daizhe Decoction.
