乙醇/磷酸氢二钾(K_(2)HPO_(4))双水相提取核桃青皮多糖工艺及其抗氧化活性的研究 被引量:3

摘要 为了探讨核桃青皮多糖的提取工艺及其抗氧化功能,以多糖提取率为考察指标,采用乙醇(ethanol)/磷酸氢二钾(K_(2)HPO_(4))双水相体系提取核桃青皮多糖,并采用响应面Box-Behnken耦合遗传算法优化双水相提取核桃青皮多糖(walnut peel polysaccharides,WPPs)工艺,采用红外光谱分析方法(FT-IR)探究了WPPs结构,并检测其体外抗氧化活性。结果显示:双水相提取WPPs的最佳工艺为K_(2)HPO_(4)质量分数16%,提取时间34 min,提取温度28℃,WPPs的提取率为111 mg/g;WPPs存在多糖物质的特征吸收峰。双水相技术制备所得WPPs可有效清除ABTS自由基。说明双水相体系可以安全、价廉、高效提取核桃青皮中的多糖,并且保留了WPPs的生物活性。 In order to explore the extraction technology and antioxidant function analysis of polysaccha-rides from walnut peel,in this study,ethanol/K_(2)HPO_(4) was used to extract polysaccharides from walnut peel.The extraction rate of polysaccharide was used as the index.On this basis,response surface Box-Behnken coupled genetic algorithm was used to optimize the two-water phase extraction process of wal-nut peel polysaccharide.The structure was characterized by Fourier Transform Infrared Spectroscopy and its antioxidant activity was studied in vitro.The results showed that the optimal extraction process of poly-saccharides from walnut peel by dual-phase technology was as follows:in the dual-phase system,the lower phase K_(2)HPO_(4) was 16%mass fraction,the dual-phase extraction time was 34 min,and the dualphase extraction temperature was 28℃,so the WPPs yield of 111 mg/g could be achieved.The polysac-charides from walnut peel had characteristic absorption peaks of polysaccharide compounds.The polysac-charides of walnut peel prepared by aqueous two-phase method has significant scavenging effect on ABTS free radicals,the two-phase aqueous system can extract polysaccharides from walnut peel security,inexpensive and effectively,and retain the biological activity of polysaccharides from walnut peel.
