摘要
为进一步掌握新疆南疆地区集约化奶牛场支原体的分子特征及耐药现状,采集8个集约化奶牛场具有呼吸道症状的犊牛鼻拭子,进行支原体分离鉴定及耐药基因检测。结果显示:牛鼻拭子样品的支原体阳性率为39.02%(16/41),共检出4类支原体,分别为牛支原体、牛鼻支原体、殊异支原体、精氨酸支原体;4类支原体与国外多个流行株同源性为92.4%~100%;经分离纯化,共获得4株牛支原体、2株殊异支原体,镜检呈无细胞壁、“煎蛋样”菌落,Dienes染色菌落呈中心深蓝色、边缘浅蓝色的透明圈,生化鉴定结果符合牛支原体、殊异支原体生化特性;牛支原体分离株TEM、gyrA、tetC基因检出率均为100%,parC检出率为75.0%,殊异支原体分离株TEM、gyrA、parC和tetC的检出率均为100%,含3种、4种耐药基因的分离株占比分别为16.67%、83.33%。本研究为该地区牛支原体病防控提供了数据支撑,为指导该地临床用药,研发牛支原体病疫苗奠定了基础。
In order to further investigate the molecular characteristics and drug resistance status of Mycoplasma in intensive dairy farms in Southern Xinjiang,nose swabs were collected from the calves with respiratory symptoms in 8 farms for isolation and identification of Mycoplasma as well as for drug resistance gene testing.The results revealed that the positive rate of Mycoplasma was 39.02%(16/41),involving 4 types,namely Mycoplasma bovis,Mycoplasma bovirhinis,Mycoplasma dispar and Mycoplasma arginini,all of which were 92.4%-100%identity to several epidemic strains abroad;after isolation and purification,a total of 4 strains of Mycoplasma bovis and 2 strains of Mycoplasma dispar were obtained.Free cell-wall and "fried egg like"colonies were observed under microscopic examination,and by Dienes staining,the colonies showed transparent circles with dark blue centers and light blue edges.Meanwhile,the biochemical identification results were consistent with the biochemical characteristics of Mycoplasma bovis and Mycoplasma dispar.All the detection rates of TEM,gyrA and tetC genes of the isolated strains of Mycoplasma bovis were 100%,that of parC was 75.0%,and all the detection rates of TEM,gyrA,parC and tetC genes of the isolated strains of Mycoplasma dispar were 1o0%,the proportions of isolates containing three or four drug resistant genes were 16.67%and 83.33%,respectively.Data support was provided for the prevention and control of bovine mycoplasmosis in the region,contributing to the guidance of clinical administration and development of vaccines of bovine mycoplasmosis.
