摘要
目的研究超氧化物歧化酶1(SOD1)在宫颈癌中的表达,并探讨其与宫颈癌患者预后的关系。方法选择新疆医科大学附属肿瘤医院2016年11月至2019年8月手术切除标本共150例,包括宫颈鳞癌、宫颈上皮内瘤变(CIN)和正常宫颈组织各50例。应用免疫组化检测SOD1在各组织中的表达,借助R2生物信息平台挖掘宫颈癌中SOD1的mRNA表达及与预后的关系。结果SOD1在宫颈鳞癌中表达最高、CIN次之、正常宫颈组织中最低,差异有统计学意义(76%vs 52%vs 32%,χ^(2)=19.50,P<0.05);在正常宫颈组织、CIN与宫颈鳞癌组织中,随着宫颈病变级别的升高,SOD1胞核阳性表达率逐渐升高,其差异具有统计学意义(χ^(2)=6.850,P<0.05);在宫颈癌组织的胞核中表达量显著升高,并与组织浸润有关(χ^(2)=10.424,P<0.05);对R2生物信息平台数据的挖掘和分析结果显示,SOD1 mRNA在宫颈癌所有临床分期中均有表达;Kaplan-Meier生存分析提示,SOD1高表达组患者有更好的预后(χ^(2)=5.18,P<0.05);SOD1在不同组织学类型宫颈癌中的表达差异具有统计学意义(F=4.84,P<0.05)。结论SOD1在浸润深的宫颈癌中表达升高,且与组织学类型有关,SOD1在宫颈癌胞核的高表达与不良预后相关。
Objective To investigate expression of SOD1 in cervical cancer and to explore its relationship with prognosis of patients with cervical cancer.Methods A total of 150 surgical resection specimens were obtained from November 2016 to August 2019 in The Affiliated Tumor Hospital of Xinjiang Medical University,including 50 cases of cervical squamous carcinoma,50 cases of cervical intraepithelial neoplasia(CIN)and 50 cases of normal cervical tissues.Immunohistochemistry was used to detect expression of SOD1 in different cervical tissues,and relationship of mRNA expression of SOD1 in cervical cancer with prognosis of the patient was explored with help of R2 bioinformatics platform.Results The expression of SOD1 was the highest in cervical squamous carcinoma tissues,the second in CIN tissues,and the lowest in normal cervical tissues,there were statistically significant differences(76%vs 52%vs 32%,χ^(2)=19.50,P<0.05).In an order of normal cervical tissues,CIN tissues and cervical squamous carcinoma tissues,along with increase in severity and grade of cervical lesions,the positive expression rate of SOD1 in nucleus elevated gradually,and the differences were significant(χ^(2)=6.850,P<0.05).And the expression amount of SOD1 in nucleus of cervical cancer cells significantly elevated and was associated with tissue infiltration(χ^(2)=10.424,P<0.05).Mining and analysis of R2 Bioinformatics Platform data showed that SOD1 mRNA was expressed in tissues of all clinical stages of cervical cancer,Kaplan-Meier survival analysis suggested that the patients in the SOD1 high-expression group had a better prognosis(χ^(2)=5.18,P<0.05).Its expression was statistically different among different histological types of cervical cancer and was the highest in cervical squamous carcinoma(F=4.84,P<0.05).Conclusion The expression of SOD1 elevates especially in deeply infiltrated cervical carcinoma,and correlates with histological type of cervical cancer.High expression of SOD1 in nucleus of cervical cancer cells correlates with poor prognosis of the patient.
