摘要
目的 探讨柴胡煎剂是否对疟疾小鼠有缓解症状的治疗作用。方法 采用约氏疟原虫17XNL虫株感染6~8周龄小鼠,制备疟疾小鼠模型。每天尾静脉采血计算小鼠红细胞疟原虫感染率。待疟疾小鼠模型红细胞感染率达到3%左右,随机将疟疾小鼠分成治疗组和对照组。治疗组使用柴胡煎剂灌胃治疗,根据柴胡煎剂的用量不同分为低剂量组(0.5 mL,含生药0.05 g)和高剂量组(0.5 mL,含生药0.10 g);对照组使用相同体积生理盐水灌胃。每天观察并记录治疗组和对照组疟疾小鼠模型红细胞疟原虫感染率的变化;随机测量所有小鼠体温变化情况。记录小鼠的生存时间,灌胃7 d后小鼠摘眼球取血对比血红蛋白含量等常见血常规指标并手术取小鼠肝脏、脾脏称重,比较3组小鼠肝重比、脾重比。结果 治疗组和对照组小鼠红细胞疟原虫感染率并无明显差异(P>0.05);对照组小鼠体温高于治疗组;治疗组小鼠生存率明显高于对照组(P<0.05);低剂量组与高剂量组在血红蛋白含量上无差异,在白细胞计数、红细胞计数、血细胞压积、血小板计数上无差异,3组小鼠脾重比无差异(均P>0.05),高剂量组小鼠肝重比明显大于低剂量组(P<0.05)。结论 在特定剂量下柴胡煎剂对约氏疟原虫并无直接杀伤作用;但在减少小鼠发热、减轻小鼠肝肿大,提升小鼠存活率方面有一定作用。
Objective To investigate whether bupleurum decoction has therapeutic effect alleviateing the symptoms of malaria on malaria mice.Methods Malaria mouse model was prepared by infecting 6-8 weeks old mice with 17XNL strain of Plasmodium yoelii.The infection rate of plasmodium in mouse red blood cells was counted by daily tail vein sampling.When the red blood cell infection rate of malaria mouse model reached about 3%,malaria mice were randomly divided into 2 treatment groups and a control group.The treatment groups were given bupleurum decoction by gavage and divided into a low dose group(0.5 mL,containing crude drug 0.05 g)and a high dose group(0.5 mL,containing crude drug 0.1 g).The control group was given 0.5 mL normal saline by intragastric administration.The change of plasmodium infection rate in erythrocyte of malaria mouse model in the treatment groups and the control group was observed and recorded every day.The temperature changes of all mice were measured randomly.The survival time of the mice was recorded.After 7 days of intragastriction,blood was taken from the eyeballs of the mice to compare the changes of common blood routine indexes such as hemoglobin content.The liver and spleen of the mice were surgically taken and weighed to compare the liver weight ratio and spleen weight ratio of the 3 groups.Results There was no significant difference in plasmodium infection rate among the treatment groups and control group(P>0.05).The body temperature of the control group was higher than that of the treatment group.The survival rate of mice in the treatment groups was significantly higher than that in the control group(P<0.05).There was no difference in hemoglobin content,white blood cell count,red blood cell count,hematocrit,and platelet count between the low dose group and the high dose group.There was no difference in spleen weight ratio among the 3 groups(all P>0.05).The liver weight ratio of the high dose group was significantly higher than that of the low dose group(P<0.05).Conclusion Bupleurum decoction has no direct killing effect on plasmodium.yoelii at specific dose.However,it has a certain effect on reducing fever,alleviating liver enlargement,and improving survival rate of mice.
