八宝景天多酚超声-酶辅助提取工艺优化及其抗氧化活性 被引量:1

Optimization of Ultrasonic-Enzymy-Assisted Extraction of Total Polyphenols from Sedum Spectabile Boreau and Its Antioxidant Activity

摘要 目的:采用响应面分析法优化八宝景天叶中总多酚的提取条件,以期获得更为经济科学的提取工艺。方法:以多酚得率为衡量指标,先通过单因素试验考察料液比、乙醇体积分数、提取时间、超声时间、超声功率、酶量、酶解时间和酶解温度等因素对提取率的影响,再用PlackettBurman(PB)试验筛选出影响显著的因素,然后借助Box-Behnken试验确定多酚提取的最佳工艺,最后通过体外的DPPH和ABTS自由基清除率评价其体外抗氧化活性。结果:PB试验证明提取时间、超声功率和纤维素酶量为影响多酚提取条件中的显著因素。响应面分析优化提取条件为:提取时间为1 h、超声功率为300 W、纤维素酶量为15.8 mg,重复3次,多酚得率平均为24.22 mg/g;抗氧化活性结果表明,景天多酚提取液可有效地清除DPPH、ABTS等自由基,清除率最高分别为82.10%和90.20%,其IC_(50)分别为0.036 mg/mL和0.021 mg/mL。结论:该优化工艺稳定可靠,具有一定实用价值。体外抗氧化试验结果表明景天多酚具有良好的抗氧化性,为其保健食品的开发提供理论依据。 Objective:Response surface methodology was used to optimize the extraction conditions of total polyphenols from leaves of Sedum spectabile Boreau in order to obtain an economic and scientific extraction process.Methods:The content of polyphenol were used as the measurement index.On the basis of single factor experiment,the effects of the solid-liquid ratio,ethanol volume fraction,extraction time,ultrasonic power,ultrasonic time,enzyme dosage,enzymolysis time and enzymolysis temperature were explored.Plackett-Burman(PB)test design was used to screen out the significant factors affecting the yield of polyphenols.Box-Behnken test design was used to optimize the extraction process of polyphenols.Finally,the antioxidant activity was evaluated by DPPH and ABTS free radical scavenging in vitro.Results:PB experiment showed that the extraction time,ultrasonic power and dosage of cellulase were significant factors affecting the extraction of polyphenols.Response surface optimization of extraction conditions were as follows:when the extraction time was 1 h,the ultrasonic power was 300 W and the dosage of cellulase was 15.8 mg,the average yield of polyphenols was 24.22 mg/g after 3 repetitions.The antioxidant activity results showed that the extract of Seduma polyphenols could effectively remove DPPH,ABTS and other free radicals,with the highest scavenging rates of 82.10%and 90.20%,respectively,and its IC_(50)were 0.036 mg/mL and 0.021 mg/mL,respectively.Conclusion:It showed that the optimized process was reliable,stable,and worth for using.The results ofin vitro antioxidant experiment indicated that the polyphenols had good antioxidant activity,which provide theoretical basis for the development of health food.
