摘要
为探究表没食子儿茶素没食子酸酯(EGCG)、柚皮素、橙皮苷的通便作用机制,将80只SD大鼠随机分为空白对照组、便秘模型组(MG)、EGCG低剂量组(LE组)、EGCG高剂量组(HE组)、柚皮素低剂量组(LN组)、柚皮素高剂量组(HN组)、EGCG与柚皮素组(EN组)、EGCG与橙皮苷组(EH组)。采用夹尾法结合洛哌丁胺制造便秘动物模型,试验组给予不同活性物灌胃,测定血浆乙酰胆碱(Ach)表达水平,检查结肠组织c-Kit mRNA、SCF mRNA、BDNF mRNA、GDNF mRNA、Cx43 mRNA表达水平。采用免疫组化法分析肌动蛋白(Arp2/3)的表达。结果表明,夹尾结合洛哌丁胺使得大鼠血浆Ach显著降低(P<0.01),结肠组织c-Kit mRNA、SCF mRNA、BDNF mRNA、GDNF mRNA、Cx43 mRNA表达水平明显减少(P<0.01)。相比较于模型组,柚皮素、EGCG、橙皮苷具备较好的改善效果,显著提高c-Kit mRNA、SCF mRNA、BDNF mRNA、GDNF mRNA、Cx43 mRNA表达水平,同时增加Ach含量(P<0.01)。免疫组化结果显示,模型组Arp2/3分布与空白对照组相比,数目减少,分布不规则, HN组、LE组、EH组的Arp2/3数目、分布与空白对照组相似。试验结果显示EGCG与柚皮素、橙皮苷能以提升神经-ICC-平滑肌整体的稳定性而缓解便秘。
The mechanism of EGCG and naringin,hesperidin in alleviating constipation was investigated.A total of 80 SD rats were randomly divided into the control group(CON),model group(MG),EGCG low-dose group(LE),EGCG high-dose group(HE),naringin low-dose group(LE),naringin high-dose group(HE),EGCG and naringin group(EN),and EGCG and hesperidin group(EH).The constipated model of rats was established by loperamide and tail clamping.The experimental groups were orally injection of EGCG and naringin,hesperidin.The levels of plasma acetylcholine were determined.The expression levels of c-Kit mRNA,SCF mRNA,BDNF mRNA,GDNF mRNA and Cx43 mRNA in colon tissue were measured.The expression level of Arp2/3 was tested by immunohistochemical method.The results showed that loperamide and tail clamping significantly reduced the expression levels of c-Kit mRNA,SCF mRNA,BDNF mRNA,GDNF mRNA and Cx43 mRNA in colon tissue(P<0.01),and significantly decreased the content of Ach(P<0.01).Compared with the control group,EGCG and naringin,hesperidin had better effects in improving the above indicators(P<0.01).The immunohistochemical results showed that the distribution of Arp2/3 in the model group decreased compared to the control group,with irregular distribution.The number and distribution of Arp2/3 in the HN group,LE group,and EH group were similar to those in the control group.Based on the above results,the results of experiments indicated that EGCG,naringenin and hesperidin could alleviate constipation by improving the overall stability of gastrointestinal tract nerve-ICC-smooth muscle.
