盆炎通络方对输卵管炎性阻塞性不孕症模型大鼠转化生长因子-β1、Ⅱ型转化生长因子-β受体和Smad3表达的影响

摘要 目的:研究盆炎通络方对输卵管炎性阻塞性不孕症(Salpingitis Obstructive Infertility,SOI)模型大鼠转化生长因子-β1(Transforming Growth Factor-β1,TGF-β1)、Ⅱ型TGF-β受体(TGF-βTypeⅡReceptor,TβRⅡ)和Smad3表达的影响,并探讨其作用机制。方法:采用双侧输卵管注射苯酚糊剂法制备SOI大鼠模型。造模成功后随机分为假手术组、模型组、康妇炎胶囊组、盆炎通络方低剂量组、盆炎通络方中剂量组、盆炎通络方高剂量组共6组。灌胃治疗30 d后,观察输卵管外观形态,并收集输卵管组织,采用苏木精-伊红染色法观察病理情况,免疫组化法检测输卵管组织中TGF-β1、TβRⅡ和Smad3表达。结果:肉眼观察,与模型组对比,各治疗组输卵管组织充血、肿胀现象均有不同程度的改善,以盆炎通络方高剂量组最为明显。光镜下,各治疗组均不程度地减轻了大鼠输卵管组织炎性反应、改善了输卵管组织纤维化。与假手术组对比,模型组输卵管组织中TGF-β1、TβRⅡ和Smad3表达量明显增加(P<0.01)。与模型组对比,各治疗组输卵组织中TGF-β1、TβRⅡ和Smad3表达量均减少(P<0.05,P<0.01)。结论:盆炎通络方是治疗SOI的有效方剂,其可能的机制是下调输卵管组织中TGF-β1及TβRⅡ和Smad3表达,减轻炎症反应、抑制输卵管组织纤维化,从而改善输卵管组织结构。 Objective:To investigate the effect of the Penyan Tongluo prescription(盆炎通络方)on levels of transforming growth factor-β1,transforming growth factor-βtypeⅡreceptor and Smad3 in salpingitis obstructive infertility(SOI)rats,and explore its action mechanism.Methods:SOI rat model was prepared by phenol paste injection into bilateral fallopian tubes.TheO successful model rats were randomly divided into 6 groups,including the shame-operation group,the model group,the low dose group,the medium dose group and the high dose group of the Penyan Tongluo prescription.After 30 days of intragastric treatment,the appearance and morphology of the oviduct were observed and the oviduct tissues were collected.HE staining was used to observe pathological changes,and immunohistochemistry was used to detect the expression of TGF-β1,TβRⅡand Smad3 in rat oviduct tissues.Results:Visual inspection:compared with the model group,the hyperemia and swelling of oviduct tissue in all treatment groups were relieved to different degrees,especially in the high dose group of the Penyan Tongluo prescription.Light microscopic inspection:all the treatment groups could reduce the inflammatory reaction and relieve the fibrosis of rat oviduct tissue in different degrees.The expression of TGF-β1,TβRⅡand Smad3 in the model group were higher than those in the shame-operation group(P<0.01).The expression of TGF-β1,TβRⅡand Smad3 in all treatment groups were lower than those in the model group(P<0.05,P<0.01).Conclusion:The Penyan Tongluo prescription is an effective prescription on SOI.The possible mechanism is to down-regulate the expression of TGF-β1,TβRⅡand Smad3,reduce the inflammatory response,inhibit the fibrosis,and thus ameliorating the structure of the tubal tissue.
