摘要
[目的]探讨促红细胞生成素(EPO)对单侧输尿管梗阻小鼠Caspase-3、α-SMA、Bcl-2蛋白表达的影响,为临床应用EPO治疗肾损伤提供实验依据.[方法]取90只2~4周龄清洁级雄性昆明小鼠,随机分为假手术组(Sham组)、肾损伤模型组(UUO组)和EPO治疗组(EPO组,每日500 U/kg).术后第3、7、14天时每组分别随机选择10只小鼠检测血清中的超氧化物歧化酶(SOD)和丙二醛(MDA)水平,采用HE染色法观察各组肾组织病理学变化,利用蛋白免疫印迹法测定肾组织中Caspase-3、α-SMA及Bcl-2蛋白表达水平.[结果]与Sham组比较,UUO组SOD水平显著降低,MDA水平显著升高,肾组织中Caspase-3、α-SMA蛋白表达水平显著升高,Bcl-2蛋白表达水平显著降低,相比较差异均有统计学意义(P<0.05);与UUO组比较,EPO组SOD水平显著升高,MDA水平显著降低,肾组织中Caspase-3、α-SMA蛋白表达水平显著降低,Bcl-2蛋白表达水平显著升高,相比较差异亦均有统计学意义(P<0.05).TUNEL染色结果显示,UUO组凋亡细胞数量较Sham组显著增多,EPO组较UUO组显著减少,差异均有统计学意义(P<0.05).[结论]EPO可改善单侧输尿管梗阻模型小鼠肾功能损伤,抑制肾组织细胞凋亡,对肾组织具有抗氧化应激作用.
OBJECTIVE To investigate the effects of erythropoietin(EPO)on the expressions of caspase-3,α-SMA and Bcl-2 proteins in mice with unilateral ureteral obstruction,and to provide experimental evidence for clinical application of EPO in the treatment of renal injury.METHODS Ninety clean male Kunming mice aged 2-4 weeks were selected and randomly divided into sham operation group(Sham group),renal injury model group(UUO group)and EPO treatment group(EPO group,500 U/kg per day).On the 3 rd,7 th and 14 th day after operation,the levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in serum of 10 mice randomly selected from each group were detected.The pathological changes of renal tissue were observed by HE staining.The expressions of caspase-3,α-SMA and Bcl-2 protein in renal tissues were determined by western blot.RESULTS Compared with the Sham group,the level of SOD was significantly decreased,and the level of MDA was significantly increased,the expressions of caspase-3 protein andα-SMA protein were significantly increased,and the Bcl-2 protein expression was significantly decreased in the UUO group,and the differences were statistically significant(P<0.05).Compared with the UUO group,the SOD level was significantly increased and the MDA level was significantly decreased,the expressions of caspase-3 protein andα-SMA protein were significantly decreased,and the expression of Bcl-2 protein was significantly increased in the EPO group,and the differences were statistically significant(P<0.05).TUNEL staining results showed that the number of apoptotic cells in the UUO group was significantly higher than that in the Sham group,and the number of apoptotic cells in the EPO group was significantly lower than that in the UUO group,and the differences were statistically significant(P<0.05).CONCLUSION EPO can improve renal function injury in mice with unilateral ureteral obstruction,inhibit apoptosis of renal tissue,and has antioxidative stress effect on renal tissue.
