摘要
本校在面向应用型人才培养的生物技术专业中开设反向点杂交检测β-地中海贫血基因类型的实验项目。从生物安全与阳性基因型多样性的角度出发,实验室将CD41-42(-TTCT)、-28(A>G)、IVS-Ⅱ-654(C>T)、CD71-72(+A)、CD17(A>T)以及野生型共6种常见的基因型克隆至T载体中,建立β-珠蛋白基因突变类型模板库,丰富实验内容。同时笔者对实践教学提出思路,首先,小教师+翻转课堂教学法充分激发学生学习探索的兴趣;其次,在课堂的实验等待时间组织讨论,充分结合理论与实验知识点,以提高教学效果;最后,思政教育与实践同行,培养科学逻辑思维与良好实验习惯,并鼓励科普宣教。
Our college has set up an experimental project to detect the gene type ofβ-thalassemia by reverse dot hybridization in the biotechnology major for the training of applied talents.From the point of view of biosafety and diversity of positive genotypes,five common genotypes,CD41-42(-TTCT)、-28(A>G)、IVS-Ⅱ-654(C>T)、CD71-72(+A)and wild type,were cloned into T vector to establish a template library ofβ-globin gene mutation types.The author also puts forward ideas on practical teaching.First of all,the little teacher and the flipped classroom teaching method can fully stimulate students'interest in learning and exploration.Secondly,organize the discussion in the waiting time of the experiment in the classroom,fully combine the theoretical and experimental knowledge points,in order to improve the teaching effect.And ideological and political education and practice exist together,cultivate scientific logical thinking and good experimental habits,and encourage popular science education.
