摘要
研究旨在调查肠道产酸克雷伯菌是否具有致病性,从新疆某规模化羊场无菌采集患病绵羊粪便样品75份进行细菌分离培养、革兰染色镜检、生化鉴定、16S rRNA基因测序,进一步进行小鼠致病试验、药物敏感试验、中药体外抑菌试验。结果显示:经分离培养、革兰氏镜检等共检出10株疑似产酸克雷伯菌,PCR扩增得到符合预期长度的条带。PCR产物测序鉴定结果显示,5株分离株为产酸克雷伯菌。产酸克雷伯菌具有一定致病性,对米诺环素、多西环素、四环素、庆大霉素、头孢哌酮、丁胺卡那、头孢曲松、呋喃唑酮、复方新诺明、氯霉素、环丙沙星、氧氟沙星高度敏感。由甘草、艾叶组成的中药方剂对产酸克雷伯菌具有抑菌作用。研究表明,患病羔羊粪便中存在产酸克雷伯菌且具有一定致病性。
The purpose of the study was to investigate the pathogenicity of Klebsiella acidogenes in the intestine.A total of 75 fecal samples of infected sheep were collected aseptically from a large-scale sheep farm in Xinjiang for bacterial isolation and culture,gram staining microscopy,biochemical identification,16S rRNA gene sequencing,and further pathogenicity test in mice,drug sensitivity test,and antibacterial test of traditional Chinese medicine in vitro.The results showed that a total of 10 suspected strains of Klebsiella acidogenes were detected by isolation,culture and Gram microscopy,and the expected length of bands was obtained by PCR amplification.PCR product sequencing results showed that five isolates were Klebsiella acidogenes.Klebsiella acidogenes was pathogenic to a certain extent,and it is highly sensitive to minocycline,doxycycline,tetracycline,gentamicin,cefoperazone,butamicana,ceftriaxone,furazolidone,cotrimoxazole,chloramphenicol,ciprofloxacin,ofloxacin.The Chinese herbal prescription composed of licorice and Folium artemisia argyi has antibacterial effect on Klebsiella acidogenes.The study indicates that Klebsiella acidogenes is present in the feces of infected lambs and has certain pathogenicity.
