摘要
为验证SmartChem 200全自动间断化学分析仪测定植物叶片中全氮、全磷的优良性,以1 a生云南松苗木为样品,建立基于SmartChem 200全自动间断化学分析仪,同时测定植物全氮和全磷的方法。结果显示:该方法测定植物中的全氮和全磷分别在0~12 mg/L和0~5 mg/L范围内呈良好线性关系,相关系数(R^(2))分别为0.9999、0.9994;经t检验测定结果与传统凯氏定氮法、钼锑抗比色法对比无显著差异;该方法与凯氏定氮法测定全氮含量呈极显著相关关系(y=0.9828 x+0.2610,R^(2)=0.9793),该方法与钼锑抗比色法所测全磷含量呈极显著相关关系(y=1.0919 x-0.1677,R^(2)=0.9667);样品的加标回收率分别是全氮98.90%~101.85%、全磷96.10%~105.20%,3个样品重复测定3次的相对标准偏差分别为2.02%~2.79%和0.25%~1.28%。研究结果表明,SmartChem 200全自动间断化学分析仪测定方法试剂量少、操作简单、结果准确可靠、回收率高,测定结果满足对植物全氮、全磷的批量检测要求。
In this paper,a method based on SmartChem200 automated chemical analyzer was developed for the simultaneous determination of total nitrogen and total phosphorus in plants using annual Pinus yunnanensis seedlings as samples in the greenhouse of Southwest Forestry University.The method showed good linearity in the range of 0-12 mg/L and 0-5 mg/L for total nitrogen and phosphorus,respectively,with correlation coefficients(R^(2))of 0.9999 and 0.9994.The results of the t-test were not significantly different from those of the traditional Kjeldahl method and molybdenum antimony colorimetric method.The recoveries of the samples were 98.90%-101.85%for total nitrogen and 96.10%-105.20%for total phosphorus,respectively.The relative standard deviations of the three samples were 2.02%-2.79%and 0.25%-1.28%,respectively.The results showed that the method is simple,easy to operate,accurate and reliable,with high recoveries,and the determination results meet the requirements for the bulk determination of total nitrogen and total phosphorus in plants.
