摘要
试验旨在探究自拟中药方制备的乳房灌注剂治疗奶牛隐性乳腺炎的抗炎作用机制。根据牛奶体细胞数(SCC)和加利福尼亚乳腺炎试验(CMT)筛选出12头患隐性乳腺炎的奶牛,随机分为4组,每组3个重复,每个重复1头牛。阳性对照组奶牛经乳腺管注射40 mL灭菌ddH2O,各治疗组奶牛每个乳区分别注射低剂量(10 mL)、中剂量(20 mL)、高剂量(40 mL)的中药乳房灌注剂,连续使用7 d。试验结束后,检测白细胞介素-1β(IL-1β)和肿瘤坏死因子-α (TNF-α)含量,应用Western blotting法检测奶牛乳腺组织中核转录因子-κB(NF-κB) p65和细胞外调节激酶(ERK1/2)、Jun N-端激酶(JNK1/2/3)、p38磷酸化水平。结果显示,中药灌注剂以剂量依赖方式降低了牛乳中SCC和乳清中IL-1β、TNF-α的含量,抑制了乳腺组织中NF-κB p65、ERK、JNK、p38的磷酸化和NLRP3炎症小体的活化。研究表明,中药灌注剂可通过抑制NF-κB、有丝分裂原活化蛋白激酶(MAPK)和NLRP3信号通路发挥治疗奶牛隐性乳腺炎作用,具有进一步开发价值。
The purpose of the study was to further explore the anti-inflammatory mechanism of breast infusion prepared by self-made Chinese prescription in the treatment of latent mastitis of dairy cows.According to milk SCC and CMT,12 cows with occult mastitis were selected and randomly divided into four groups,with three replicates in each group and one cow in each replicate.Cows in positive control group were injected 4O mL sterilized ddH,O through mammary duct,and cows in each treatment group were injected low dose(10 mL),medium dose(20 mL)and high dose(40 mL)of TCM breast infusion in each milk area,respectively,and used continuously for 7 d.After the experiment,the content of IL-1βand TNF-αwere detected,the phosphorylation levels of NF-kB p65,ERK1/2,JNK1/2 and p38 in breast tissue of dairy cows were detected by Western blotting.The results showed that the infusion reduced the content of IL-1βand TNF-αin whey and SCC in milk in a dose-dependent manner,and inhibited the phosphorylation of NF-kB p65,ERK,JNK,p38 and the activation of NLRP3 inflammatorsome in breast tissue.The study indicates that Chinese medicine perfusion agent can inhibit NF-kB,MAPK and NLRP3 signaling pathway in the treatment of latent mastitis of dairy cows,and has a value for further development.
