‘双优’葡萄组培苗根系发生的研究 被引量:1

摘要 为探究‘双优’葡萄的生根机理,筛选出较为合适的生根培养基类型,采用正交试验设计,以不同浓度IBA、NAA和蔗糖进行处理,分别于培养20、30、40 d时调查生根情况,并测定根系中的生长素(IAA)含量;通过qRT-PCR进一步分析IAA合成和信号转导相关基因的差异表达。结果表明,1/2B5培养基较为适合‘双优’葡萄生根;在生根处理期间,IAA含量总体呈上升趋势,与主根数趋势相近,而IAA合成和信号转导相关基因的表达影响IAA含量与信号转导过程,进而影响根系发育。本研究揭示了不同处理下‘双优’葡萄生根过程中生长素合成和信号转导相关基因的差异表达及生长素含量的变化规律,为促进该品种组培苗生根提供参考。 In order to explore the rooting mechanism of'Shuangyou'grape,a suitable rooting medium type was first selected,and then an orthogonal experimental design was used to investigate the rooting situation at 20,30,40 d of rooting treatment with different concentrations of IBA,NAA and sucrose,and to determine the content of IAA in roots.The root samples were assayed for the auxin(IAA)contents,and the differential expression of IAA synthesis and signal transduction-related genes was analyzed by qRT-PCR.The results showed that 1/2B5 medium was more suitable for'Shuangyou'grape rooting.During the rooting period,IAA contents generally showed an upward trend,which was similar to the trend of the number of taproots.The expression of IAA synthesis and signal transductionrelated genes affected IAA content and IAA signal transduction process,which in turn affected root development.This study revealed the differential expression of genes related to auxin synthesis and signal transduction and the changes in IAA content during rooting of'Shuangyou'grape under different treatments,in order to provide theoretical and practical guidance for rooting of'Shuangyou'grapevine tissue culture seedlings.
