两种半夏根部致病菌的比较研究 被引量:2

Comparative study on two pathogens of Pinellia root diseases

摘要 2018年春夏,贵州省遵义市发生三叶半夏(Pinellia ternata)根部病害,叶柄以上部位倒伏,且连片快速发生。为明确其病原菌,对自然发病的半夏病株进行了病原菌的分离,并对获得的主要病原菌进行形态学观察、16S rDNA序列鉴定和致病性测定。结果表明,当地发生的半夏病害主要由2种病原菌引起:(1)胡萝卜果胶杆菌(Pectobacterium carotovorum)引起半夏叶柄以上部位水渍状快速软烂倒伏,伴随发臭;(2)铜绿假单胞菌(Pseudomonas aeruginosa)引起半夏叶柄基部折断,植株干枯黄化,连片倒伏,但单株发病进程相对较慢。这是首次有关铜绿假单胞菌引起半夏根部病害的报道,建议将其引起的病害称之为“半夏黄化猝倒病”。在人工接种条件下,铜绿假单胞菌YD-01寄主范围较窄,但能侵染番茄果实;而胡萝卜果胶杆菌RD-02可以侵染莴苣、青椒、茄子、土豆、番茄和梨等果实,并产生明显软腐症状。利用BioF型间歇浸没植物生物反应器建立植物-微生物共培养模型,结合盆栽试验分析2种病原菌导致的病情指数和病程指标变化,进一步确定了其差异。本研究结果为半夏根部病害防控提供了新的直接依据。 Root disease of Pinellia ternata was noticed in Zunyi,Guizhou Province,during the spring and summer of 2018,leading to typical symptom like lodging above petioles,and such disease could rapidly spread across the growing fields.Thus,the causing pathogens were isolated,and characterized by morphological observation and 16S rDNA sequence analysis,together with pathogenic tests.Two major pathogens,Pectobacterium carotovorum and Pseudomonas aeruginosa,were identified for the disease of Pinellia.The former resulted in watery soft rot above petioles,subsequently with plant falling down and bad smell,while the latter caused Pinellia petioles to break down,then the plants turned dry and yellowing,ultimately fell down in large area,despite relatively show development of this disease.To the best of our knowledge,this is the first report which demonstrated P.aeruginosa could cause Pinellia disease,so we would accordingly name this new disease as “Pinellia yellow damping-off disease”.Under artificial inoculation conditions,P.aeruginosa YD-01 could only infect a narrow range of hosts including Solanum lycopersicum fruits;while P.carotovorum RD-02 could infect multiple products,such as Lactuca sativa,Capsicum annuum,Solanum melongena,Solanum tuberosum,Solanum lycopersicum and Pyrus,causing obvious soft rot symptoms.Regarding the difference in terms of disease index and disease development,the two kinds of pathogenic bacteria were further investigated through a plant/microbe co-cultivation model established from a BioF intermittent immersion plant bioreactor,tegether with pot tests.Our research results provide new scientific basis for the control of Pinellia root disease.
