pH对2种外生菌根真菌生长及主要酶活性的影响 被引量:2

Effects of pH on growth and main enzyme activities of two ectomycorrhizal mycorrhizal fungi

摘要 以厚环乳牛肝菌和点柄乳牛肝菌为研究对象,采用MMN固体和液体培养基,设置pH单因子5~6区间内梯度条件,测定2种外生菌根真菌的菌丝生长速率、菌丝干重、纤维素酶和漆酶的变化特征,探究2种外生菌根真菌培养扩繁条件。结果表明:厚环乳牛肝菌在pH值5.2~5.4的MMN培养基中菌丝生长速度最快、菌丝干重较高,点柄乳牛肝菌在pH 5.8的MMN培养基中菌丝生长速度最快、菌丝干重较高。在不同pH处理的MMN中,点柄乳牛肝菌的纤维素酶和漆酶活性高于厚环乳牛肝菌中的2种酶活性,在pH 5.4的MMN中厚环乳牛肝菌的纤维素酶活性最高为0.694 U·mL^(-1)。在pH 5.2的MMN中厚环乳牛肝菌的漆酶活性最高为0.164 U·mL^(-1)。在pH 5.8的MMN中,点柄乳牛肝菌的纤维素酶和漆酶活性最高分别为1.518、0.715 U·mL^(-1)。结合菌丝生长情况与2种酶活性变化可知,pH 5.2~5.4的MMN更适合厚环乳牛肝菌的培养,pH 5.8的MMN更适合点柄乳牛肝菌的培养。 Suillus grevillea and Suillus granulatus were the research objects in this experiment, MMN solid and liquid medium were used to set the gradient conditions within the range of pH single factor 5-6, and the mycelial growth rate, dry weight of mycelium, cellulase and laccase of the two ectomycorrhizal fungi were measured, to provide technical reference for the culture and propagation of two ectomycorrhizal fungi and the mycorrhizal seedlings of Pinaceae. The results showed that: The growth rate of mycelium and the dry weight of Suillus grevillea was the fastest and higer in the MMN of pH 5.2-5.4. The growth rate of mycelium and the dry weight of Suillus grevillea was the fastest and higer in the MMN of pH 5.8. The cellulase and laccase activities of Suillus granulatus were higher than those of Suillus grevillea In MMN treated with different pH. The cellulase of Suillus grevillea was higher in the pH 5.4 of the MMN, the maximum of cellulase activity respectively was 0.694 U·mL^(-1). The highset laccase activity of Suillus grevilleais in the pH 5.2 of the MMN, was 0.164 U·mL^(-1). The cellulase and laccase activity of Suillus granulatus were highest in the pH 5.8 of the MMN, the maximum of cellulase and laccase activity respectively was 1.518 and 0.715 U·mL^(-1). Combined with the growth of mycelium and the changes of the two enzyme activities, MMN with pH 5.2~5.4 was more suitable for the cultivation of Suillus grevillea;MMN with pH 5.8 was more suitable for the cultivation of Suillus granulatus.
