基于网络药理学研究丹栀调脂汤改善代谢相关脂肪性肝病的作用机制 被引量:1

摘要 目的:使用网络药理学方法探究丹栀调脂汤对于改善代谢相关脂肪性肝病的作用机制。方法:通过TCMSP数据库获得丹栀调脂汤的药物成分并通过Gene Cards,OMIM,Pharmgkb,TTD和DisGenet数据库查找疾病相关靶点,利用Cytoscape软件建立丹栀调脂汤的单味药-组分-作用靶点,使用STRING绘制PPI网络。运用GO和KEGG富集分析对所得到的所有靶点进行系统分析。结果:丹栀调脂汤治疗胰岛素抵抗潜在作用靶点有106个,包括AKT1、IL-6、TNF、JUN、MAPK8、STAT3、VEGFA等。丹栀调脂汤作用的信号通路涉及157条,包括PI3K-Akt信号通路、MAPK信号通路、TNF通路等。结论:丹栀调脂汤在改善胰岛素抵抗方面可能有多种途径,能够通过减轻肝脏纤维化,延缓代谢相关脂肪性肝病的发生发展。 Objective:To explore the mechanism of Danzhi Tiaozhi Decoction to improve metabolism-related fatty liver disease through network pharmacology.Methods:The TCMSP database was used to obtain the drug components of Danzhi Tiaozhi Decoction and to identify disease-related targets through Gene Cards,OMIM,Pharmgkb,TTD and DisGenet databases.Cytoscape software was used to establish the single drug-component-target of Danzhi Tiaozhi Decoction,and STRING was used to draw the PPI network.All the targets were systematically analyzed by GO and KEGG enrichment analysis.Results:There were 106 potential targets for the treatment of insulin resistance by Danzhi Tiaozhi Decoction,including AKT1,IL-6,TNF,JUN,MAPK8,STAT3,VEGFA,etc.There were 157 signaling pathways involved in Danzhi Tiaozhi Decoction,including PI3K-Akt signaling pathway,MAPK signaling pathway,TNF signaling pathway,etc.Conclusion:Danzhi Tiaozhi Decoction may have many ways to improve insulin resistance,which can reduce liver fibrosis and delay the occurrence and development of metabolism-related fatty liver disease.
