奶牛隐性乳房炎抗性相关基因多态性与体细胞评分关联分析 被引量:4

Association analysis of recessive mastitis resistance-related gene polymorphisms with somatic cell scores in dairy cows

摘要 为了探讨中国荷斯坦奶牛隐性乳房炎相关基因NOD2c.4500、CXCR1+777、SPP1-1301、 TLR4+2021、CXCR2+777、 BRCA1c.46126单核苷酸多态性(SNPs)位点与体细胞评分的关联性,试验利用PCR-SSCP及Sanger测序技术对135头中国荷斯坦奶牛的6个SNPs位点进行基因分型,分析SNPs位点与中国荷斯坦奶牛体细胞评分的相关性,并运用多座位外显方差分析法(MPVA)对不同多基因聚合与奶牛体细胞数的相关性进行研究。结果表明:在6个SNPs位点上均检测到碱基突变,在NOD2c.4500 C>A、CXCR1+777 G>C、SPP1-1301 G>A、TLR4+2021 C>T位点上均检测到3种基因型,其中优势基因型分别为CA、CC、AA、CC,优势等位基因分别为A、G、A、C。在CXCR2+777 G>C、BRCA1c.46126 G>T位点上均检测到2种基因型,其中优势基因型均为GG,优势等位基因均为G。SPP1-1301 G>A、TLR4+2021 C>T位点均处于低度多态(PIC<0.25),其余位点则均处于中度多态(0.25<PIC<0.5);6个SNPs位点均处于Hardy-Weinberg平衡状态(P>0.05)。CXCR2+777 G>C、TLR4+2021 C>T位点与体细胞评分的关联未达到显著差异(P>0.05);NOD2c.4500 C>A位点CA基因型、CXCR1+777 G>C位点CC基因型、SPP1-1301 G>A位点AA基因型、BRCA1c.46126 G>T位点GT基因型个体的体细胞评分显著低于其他基因型个体(P<0.05)。SPP1-1301-CXCR1+777-NOD2 c.4500为多基因聚合分析中最佳多座位组合,SPP1(AA)-CXCR1(CC)-NOD2(CA)为最佳基因型组合。说明SPP1-1301、CXCR1+777、NOD2c.4500位点与中国荷斯坦奶牛体细胞评分显著相关,可作为奶牛隐性乳房炎抗性基因的分子辅助标记。 In order to investigate the association between single nucleotide polymorphisms(SNPs) loci of recessive mastitis-associated genes NOD2c.4500, CXCR1+777, SPP1-1301, TLR4+2021, CXCR2+777, BRCA1c.46126, and somatic cell scores in Chinese Holstein cows, the experiment used PCR-SSCP and Sanger sequencing techniques to genotype six SNPs loci in 135 Chinese Holstein cows, to analyze the correlation with somatic cell scores in Chinese Holstein cows, and to use multi-seat epistatic variance analysis(MPVA) to investigate the association between different multigene polymerization and somatic cell scores in cows. The results showed that base mutations were detected at all six SNPs loci, and three genotypes were detected at loci NOD2c.4500 C>A, CXCR1+777 G>C, SPP1-1301 G>A, and TLR4+2021 C>T. Among them, the dominant genotypes were CA, CC, AA, and CC, and the dominant alleles were A, G, A, and C, respectively. Two genotypes were detected at both loci CXCR2+777 G>C and BRCA1c.46126 G>T, with GG being the dominant genotype and both dominant alleles being G. The loci SPP1-1301 G>A and TLR4+ 2021 C>T were all in low polymorphism(PIC<0.25), while the remaining loci were all in moderate polymorphism(0.250.05). CXCR2+777 G>C and TLR4+2021 C>T loci did not reach significant difference in association with somatic cell scores(P>0.05);individual somatic cell scores of NOD2c.4500 C>A locus CA genotype, CXCR1+777 G>C locus CC genotype, SPP1-1301 G>A locus AA genotype, and BRCA1c.46126 G>T locus GT genotype were significantly lower than those of individuals with other genotypes(P<0.05). SPP1-1301-CXCR1+777-NOD2 c.4500 was the best multi-seat combination in multigene polymerization analysis, and SPP1(AA)-CXCR1(CC)-NOD2(CA) was the best genotype association. The results suggested that SPP1-1301, CXCR1+777, and NOD2c.4500 loci were significantly associated with somatic cell scores in Chinese Holstein cows and could be used as molecular auxiliary markers for recessive mastitis resistance genes in cows.
