摘要
为比较虾原肌球蛋白和卵清蛋白分别诱导的小鼠过敏性哮喘肺上皮损伤,以同等剂量的虾原肌球蛋白(虾原肌球蛋白、虾原肌球蛋白含佐剂)或卵清蛋白(卵清蛋白、卵清蛋白含佐剂)每周腹腔注射致敏小鼠1次,连续3周,再进行持续7 d的抗原滴鼻激发诱导小鼠过敏性哮喘模型。测定肺指数,采用天狼星红染色法观察肺组织嗜酸性粒细胞浸润情况;PAS染色法观察肺组织支气管黏膜上皮增生和分泌黏液情况;qRT-PCR法检测肺组织上皮源性相关因子Tslp和IL-25,以及上皮紧密连接蛋白Zo-1、Ocln和Cldn18的mRNA表达水平。结果表明:与对照组相比,虾原肌球蛋白组(虾原肌球蛋白、虾原肌球蛋白含佐剂)、卵清蛋白含佐剂组均能诱导出明显的肺指数升高,肺组织嗜酸性粒细胞浸润和支气管黏膜细胞高分泌,肺组织上皮源性相关因子Tslp和Il-25的mRNA水平上调,上皮屏障紧密连接蛋白Zo-1、Ocln和Cldn18表达下调,但卵清蛋白组未见明显改变。在含佐剂组,虾原肌球蛋白组与卵清蛋白组相比,其诱导的嗜酸性粒细胞炎症,肺组织Tslp和Il-25 mRNA水平上调和Zo-1 mRNA水平下调更明显。这一研究提示虾原肌球蛋白能诱导更强的小鼠过敏性哮喘肺上皮损伤。
To compare the lung epithelial injury in allergic asthma mice induced by shrimp tropomyosin and ovalbumin. The same dose of shrimp tropomyosin and ovalbumin was used for the sensitization of mice by intraperitoneal injection, with or without adjuvant once a week for three consecutive weeks, followed by a nasal challenge for seven days to induce the allergic asthma model in mice. Lung index was measured. Sirius Red staining was used for evaluating the degree of eosinophil inflammation. The hyperplasia and mucus secreting of bronchial mucosa epithelium in lung tissue were observed by PAS staining. The levels of the mRNA expression of epithelial-derived cytokines(Tslp and Il-25) and tight junction proteins(Zo-1, Ocln and Cldn18) in lung tissues were determined by qRT-PCR. The results showed that compared with the control group, in the groups including shrimp tropomyosin with or without adjuvant and ovalbumin with adjuvant induced eosinophil infiltration, mucosa hypersecretion, the higher mRNA expression levels of Tslp and Il-25, and the lower mRNA expression levels of Zo-1, Ocln and Cldn18 in lung tissue, while these responses were not observed in mice of ovalbumin sensitized mice without adjuvant. In the condition of adjuvant treatment, eosinophil inflammation and the mRNA expression levels of Tslp, Il-25 and Zo-1 in lung tissue were higher in mice group of shrimp tropomyosin plus adjuvant than those in ovalbumin-sensitized group with adjuvant. This research suggested that shrimp tropomyosin induces a stronger lung epithelial injury in allergic asthma.
