摘要
                
                    以虾原肌球蛋白每周腹腔注射致敏小鼠1次,连续3周,再持续进行6周滴鼻激发诱导小鼠发生气道重塑。采用ELISA法检测血清总IgE水平,测定肺气道阻力,HE染色观察肺组织形态学变化,PAS染色观察气道上皮细胞黏液分泌状态,Masson三色法检测胶原纤维增生,免疫组织化学法检测肺组织α-平滑肌肌动蛋白的表达,以探索建立一种以虾原肌球蛋白为致敏原诱导的小鼠气道重塑模型。结果表明:与对照组相比,血清总IgE水平明显升高,气道阻力升高,肺组织出现明显的结构性改变,表现为细支气管周炎症细胞浸润,气道壁增厚,黏液高分泌,肺间质和气道壁胶原沉积,细支气管平滑肌肥大(α-平滑肌肌动蛋白阳性面积增加)。结果说明虾原肌球蛋白成功诱导小鼠气道重塑。
                
                Mice were sensitized with intraperitoneal injections of ST once weekly for 3 consecutive weeks, followed by intranasal challenge for 6 consecutive weeks to induce airway remodeling. The serum total IgE levels were measured by ELISA. Lung airway resistance was measured. Lung tissue morphology changes were observed by HE staining. Airway epithelial mucus secretion was observed by PAS staining. Collagen deposition was detected by Masson’s trichrome stain. The expression of α-SMA was measured by immunohistochemistry stain. To develop murine models of airway remodeling induced by ST as an allergen. The model mice showed elevated IgE levels in sera and increased lung airway resistance. Compared with the control group, severe structural changes in lung tissue were observed in the model group, characterized by the inflammatory cell infiltration, airway wall thickening, mucus hypersecretion, collagen deposition in the interstitial and airway wall, and an increase in the area of peribronchial smooth muscle as assessed by immunostaining with an anti α-smooth muscle actin Ab. Conclusion Airway remodeling is successfully induced by ST in mice.
