口蹄疫O型、A型二价3B表位缺失灭活疫苗在奶水牛的免疫效果观察 被引量:1

Observation on Immune Efficacy of Inactivated Vaccine with Bivalent 3B Epitope Deletion of Foot-and-Mouth Disease Type O and A in Dairy Buffalo

摘要 为了评价口蹄疫(FMD)O型、A型二价3B表位缺失灭活疫苗在奶水牛临床应用的安全性及免疫效果,本试验选择90~100日龄健康奶水牛犊牛24头,平均分为2个组,试验组接种表位缺失疫苗,在首免后30 d,二免后180 d加强免疫,对照组按照相同的免疫程序接种常规疫苗;在不同时间点采集血清进行口蹄疫O型、A型抗体效价和非结构蛋白3ABC抗体检测,采集食道-咽分泌液(OP)进行口蹄疫病原学检测。结果显示:在抗体平均滴度方面,试验组抗体比对照组高1.4~2.8个滴度,在9个免疫后时间点试验组O型抗体平均滴度全部显著(P<0.05)甚至极显著(P<0.01)高于对照组,7个免疫后时间点试验组A型抗体平均滴度显著(P<0.05)甚至极显著(P<0.01)高于对照组;试验组抗体合格率比对照组高出25%~41%;口蹄疫非结构蛋白3ABC抗体标记疫苗免疫动物均为阴性。综合分析表明,口蹄疫O型、A型二价3B表位缺失灭活疫苗具有良好的免疫保护效果,在感染抗体水平上也能保证非结构蛋白抗体阴性。本试验为口蹄疫净化和口蹄疫无疫小区、无疫区建设探索可行的方法。 To evaluate the safety and immune effect of inactivated vaccine with bivalent 3B epitope deletion of foot-and-mouth disease(FMD)type O and A in dairy buffaloes,this study divided 24 healthy 90-100-day-old dairy calves in two equal groups.The test group was immunized with epitope deficient vaccine at days 0,30 and 180,while the control group was immunized with routine vaccine using the same schedule.Sera were collected at different times to detect the titers of type O and A antibodies and 3ABC antibodies to non-structural proteins of FMD,and oesophagus-pharyngeal secretions(OP)were collected for etiological detection of FMD.The results showed that the average antibody titers in the test group was 1.4-2.8 higher than those in the control group;the average type O antibody titers in the test group were significantly(P<0.05)or even highly significantly(P<0.01)higher than those in the control group at 9 time points after immunization;the average type A antibody titers in the test group were significantly(P<0.05)or even highly significantly(P<0.01)higher than those in the control group at 7 time points after immunization.The qualified antibody rates in the test group were 25%-41%higher than those in the control group.The dairy buffaloes immunized with 3ABC antibody labeled non-structural protein of FMD were negative.Comprehensive analysis showed that the inactivated vaccine with bivalent 3B epitope deletion of FMD type O and A had good immune protection effect and could ensure negative antibody against non-structural protein at the level of infection antibody.This study presents a feasible method for the cleansing of foot-and-mouth disease and for the establishment of an area free of foot-and-mouth disease.
