基于网络药理学的半夏麻黄丸抗心律失常作用机制研究 被引量:3

摘要 目的利用网络药理学方法探讨半夏麻黄丸治疗心律失常的潜在作用机制。方法借助中药系统药理学数据库和分析平台(TCMSP),以类药性(DL)、脂水分配系数对数值(ALogP)为条件筛选半夏麻黄丸的活性成分并收集靶点信息;使用人类基因数据库(Genecard)、人类孟德尔遗传数据库(OMIM)获取疾病靶点,进而构建韦恩图,得到半夏麻黄丸治疗心律失常的潜在作用靶点;采用Cystoscap 3.7.2软件构建“中药-成分-疾病-靶点”网络,运用STRING数据库构建蛋白互相作用(PPI)网络,并对靶点基因功能(GO)和KEGG通路进行分析。结果从半夏麻黄丸中共筛选出槲皮素、芹菜素、木犀草素等36个活性成分,涉及包括PTGS2、PRSS1、RXRA、ADRB2、DPP4在内的165个基因靶点,主要富集于2467条生物过程、155条分子功能和89个细胞组分,调节175条与心律失常相关的信号通路。结论半夏麻黄丸可能是通过CRP、CXCL2、CCL2等靶点,调控PI3K-Akt、TNF、IL-17等信号通路,从而发挥多成分、多靶点和多通路治疗心律失常的作用优势。 Objective To explore the potential mechanism of Banxia Mahuang Pills in treating arrhythmias by network pharmacology.Methods TCMSP was used to screen the active components and collect the target information of Banxia Mahuang Pills by drug-like(DL)and lipid-water distribution coefficient logarithmic(ALog P).The databases of Genecard,OMIM were searched to obtain the disease targets.The Wayne diagram was constructed to obtain the potential targets of Banxia Mahuang Pills in the treatment of arrhythmias.The herb-components-disease-targets network was constructed by using Cystoscap3.7.2 software,and STRING database was used to constructed the PPI network constructed by,and to analyze the gene ontology(GO)and KEGG pathway.Results A total of 36active components such as Quercetin,Apigenin and Luteolin were selected from Banxia Mahuang Pills,which can act on 165 target genes such as PTGS2、PRSS1、RXRA、ADRB2 and DPP4.The targets were mailly enriched on 2467 biological processes,155 molecular functions and 89 cell coomponents.175 signal pathways were related to the regulation of arrhythmia.Conclusion Banxia Mahuang Pills may regulate PI3K-Akt,TNF,IL-17 and other signal pathways through CRP,CXCL2,CCL2 and other targets,thus giving full play to the advantages of multi-component,multi-target and multi-pathway in the treatment of arrhythmias.
