基于网络药理学归肾丸治疗卵巢储备功能下降作用机制探讨 被引量:2

Mechanism of guishen pill on diminished ovarian reserve based on network pharmacology

摘要 目的基于网络药理学和分子对接法探讨归肾丸治疗卵巢储备功能下降(DOR)的作用机制。方法中药系统药理数据库和分析平台(TCMSP)中获取归肾丸有效成分。TCMSP及Swiss Target Prediction数据库获取有效成分相关靶蛋白,GeneCards、OMIM数据库获得DOR的相关靶蛋白,将药物靶点、疾病靶点取交集并采用RStudio软件绘制韦恩图,筛选归肾丸治疗DOR的潜在作用靶蛋白。应用STRING 11.0平台及Cytoscape软件构建蛋白-蛋白相互作用网络;应用Cytoscape构建中药-有效成分-靶点-疾病关系网络图;通过David数据库对归肾丸靶点进行GO富集分析及KEGG富集分析。结果获得归肾丸符合条件的有效化学成分共116个,参与调控DOR的潜在蛋白靶点共286个。GO富集分析共得到998个条目,其中生物过程(BP)条目783个(78%),细胞组成(CC)条目76个(8%),分析功能(MF)条目139个(14%);KEGG富集分析共获取123条通路,其中PI3K-Akt信号通路、mTOR信号通路、p53信号通路、TGF-β信号通路、胰岛素信号通路及VEGF信号通路可能为归肾丸作用的主要通路。结论归肾丸通过多成分、多靶点、多通路改善卵巢功能,调控生殖内分泌,为后续研究奠基一定基础。 Objective To explore the mechanism of Guishen Pill in treating diminished ovarian reserve based on network pharmacology.Methods The effective chemical components of Guishen pPill were obtained from the Traditional Chinese Medicine systems pharmacology database and analysis platform(TCMSP),and the TCMSP and Swiss Target Prediction databases were used to obtain the target proteins related to the effective components.GeneCards and OMIM databases were used to obtain relevant target proteins with diminished ovarian reserve function,and then,drug targets and disease targets were taken the intersection and a Venn diagram was drawn by RStudio software to screen the potential target proteins of Guishen pPill in treating diminished ovarian reserve.The STRING 11.0 platform and Cytoscape software were used to construct the protein-protein interaction network.Cytoscape was used to construct the network diagram of traditional Chinese medicines-components-targets-disease relationship.GO enrichment analysis and KEGG enrichment analysis were performed on the targets of Guishen Pill through David database.Results A total of 116 effective chemical components were obtained in accordance with the five principles of Guishen Pill,and 286 potential protein targets were involved in regulating the diminished ovarian reserve.A total of 998 items were obtained by GO enrichment analysis,including 783(78%)items for biological process(BP),76(8%)items for cell composition(CC),and 139(14%)items for analytical function(MF).A total of 123 pathways were obtained by KEGG enrichment analysis,among which the PI3 k-Akt signaling pathway,mTOR signaling pathway,p53signaling pathway,TGF-βsignaling pathway,insulin signaling pathway and VEGF signaling pathway may be the main pathways of Guishen Pill.Conclusion Guishen Pill improves ovarian function and regulates reproductive endocrinology through multiple components,multiple targets and multiple pathways.
