Study on the Correlation Between Intestinal Microcirculation and Slow Transit Constipation

摘要 目的:探讨肠微循环与慢传输型便秘(STC)的相关性。方法:检索获取STC的相关靶点,筛选其中的核心靶点进行富集分析。利用盐酸洛哌丁胺灌胃法复制STC大鼠模型,测定模型大鼠肠道碳末推进率、粪便干重、结肠黏膜微循环水平,以及血清一氧化氮(NO)和内皮素1(ET1)水平。结果:共筛选获得STC相关靶点170个,PIK3R1、PIK3CA、TP53、STAT3、AKT1等靶点可能是STC的核心靶点。富集分析结果表明,血管内皮生长因子(VEGF)信号通路可能是STC的关键信号通路。与对照组相比,模型组大鼠粪便干重低,碳末推进距离和碳末推进率小,结肠血流灌注量少,血流速度慢,血清NO水平低,ET1水平高,差异均有统计学意义,P <0.05或P <0.001。结论:STC与肠道微循环具有一定的相关性。TP53和STAT3可能是STC潜在的作用靶点,VEGF信号通路可能是STC的关键信号通路。 Objective To investigate the correlation between intestinal microcirculation and slow transit constipation. Methods The relevant targets of slow transit constipation were searched and the core targets were selected for enrichment analysis.The rat model of slow transit constipation was replicated by loperamide hydrochloride gavage,and the rat intestinal carbon particle propulsion rate,fecal dry weight,colonic mucosal microcirculation level and serum nitric oxide(NO)and endothelin 1(ET1)levels were measured. Results A total of 170 targets related to slow transit constipation were obtained,PIK3R1,PIK3CA,TP53,STAT3,AKT1 may be the core targets of slow transit constipation.The results of enrichment analysis indicated that the vaso-endothelium growth factor(VEGF)signaling pathway may be the key one of slow transit constipation.Compared with the control group,the model rats had lower fecal dry weight,smaller carbon particle propulsion distance and propulsion rate,reduced colonic blood perfusion,slower blood flow,lower serum NO level,and higher serum ET1 level,all the differences were statistically significant(P <0.05 or P <0.001). Conclusion Slow transit constipation is correlated with intestinal microcirculation.TP53 and STAT3 may be potential targets for slow transit constipation,and VEGF signaling pathway may be a key one for slow transit constipation.
