基于高效液相色谱法对淡豆豉发酵过程中6种异黄酮含量的动态分析 被引量:4

摘要 目的 分析淡豆豉发酵过程中6种异黄酮的含量动态变化,探讨淡豆豉不同发酵阶段的变化。方法 收集淡豆豉发酵过程不同阶段的样品,采用Welch Ultimate XB-C_(18)色谱柱(4.6 mm×250 mm,5 μm),以甲醇-0.1%甲酸为流动相进行梯度洗脱,检测波长为254 nm,柱温为30℃,流速为0.8 mL·min^(-1)。采用高效液相色谱法,以大豆苷、黄豆黄苷、染料木苷、大豆苷元、黄豆黄素、染料木素为对照,对淡豆豉不同发酵阶段的6种异黄酮进行含量测定。结果 淡豆豉发酵过程中,大豆苷元等3种苷元的含量总体呈上升趋势,染料木苷等3种苷的含量总体呈下降趋势。结果提示,在淡豆豉发酵过程中存在苷到苷元的转化,并且在第二次闷制阶段对于苷元含量的积累有显著影响。结论 淡豆豉发酵过程中,闷制这一环节有利于有效成分的转化和累积,第二次闷制环节是非常有必要的。 Objective To determine the dynamic change of 6 isoflavones in the fermentation of Sojae Seman Praeparatum,and the change in different fermentation stages.Methods Samples from different stages of the fermentation of Sojae Seman Praeparatum were collected.Welch Ultimate XB-C_(18) chromatographic column (4.6 mm×250 mm,5 μm) was used.The gradient elution was conducted out with methanol-0.1% formic acid as the mobile phase.The detection wavelength was 254 nm,the column temperature was 30℃,and the flow rate was 0.8 mL·min^(-1).The content of 6 isoflavones in different fermentation stages of Sojae Seman Praeparatum were determined by HPLC,with daidzin,glycitin,genistin,daidzein,glycitein,and genistein as the reference substances.Results During the fermentation of Sojae Seman Praeparatum,the content of 3 aglycones such as daidzein tended to increase while the content of three glycosides such as genistin tended to decrease.There was a conversion from glycoside to aglycone in the fermentation of Sojae Seman Praeparatum which significantly affected the accumulation of aglycone content in the second stuffy stage.Conclusion In the fermentation process of Sojae Seman Praeparatum,the process of suffocation is conducive to the transformation and accumulation of effective components,and the second suffocation process is very necessary.
