摘要
为建立多花黄精高效再生体系,试验以多花黄精种子无菌萌发后形成的幼芽为材料,采用正交试验设计,筛选其愈伤组织和不定芽诱导的较优配方,并进一步开展了不定芽增殖、生根和移栽驯化试验。结果表明,MS+6-BA 2.0 mg/L+NAA 0.5 mg/L+2,4-D 1.0 mg/L有利于诱导无菌幼芽形成愈伤组织,诱导率达83.5%;不定芽诱导较好的培养基配方为MS+6-BA 3.0 mg/L+NAA 0.5 mg/L+2,4-D 0.5 mg/L和MS+6-BA3.0 mg/L+NAA 1.0 mg/L,诱导率均为100%;不定芽增殖较好的培养基配方为MS+6-BA 2.0 mg/L+NAA0.2 mg/L,增殖系数达4.7;适宜的生根培养基配方为1/2MS+NAA 0.1 mg/L;组培苗移栽到蛭石∶泥炭土∶珍珠岩=1∶2∶1的混合基质中,成活率达100%。试验构建了以多花黄精种子为外植体的离体高效再生体系,为多花黄精种苗快速繁殖提供技术途径。
The sterile buds formed by the seeds of Polygonatum cyrtonema Hua were used as explant for establishment of efficient regeneration system,the optimal formula of callus and adventitious bud induction was selected by orthogonal design,and the adventitious bud proliferation,rooting and transplanting were further studied.The results showed that the optimal medium for callus induction was MS+6-BA 2.0 mg/L+NAA 0.5 mg/L+2,4-D 1.0 mg/L,and the induction rate was 83.5%;the optimal medium for adventitious bud induction was MS+6-BA 3.0 mg/L+NAA0.5 mg/L+2,4-D 0.5 mg/L and MS+6-BA 3.0 mg/L+NAA 1.0 mg/L,the induction rate was up to 100%.The optimal medium for adventitious bud proliferation was MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,and the multiplication coefficient was up to 4.7;the suitable medium for rooting was 1/2MS+NAA 0.1 mg/L;and the seedlings were transplanted to a mixed matrix of vermiculite∶peat∶perlite=1∶2∶1,the survival rate was up to 100%.Above all,in this experiment the efficient regeneration system was established by using the seeds of P.cyrtonema Hua,and the results provided the technical support for the rapid propagation of the P.cyrtonema.
