不同植物生长调节剂对多花黄精组织培养的效果 被引量:17

Effect of Plant Growth Regulators on Tissue Culture of Polygonatum cyrtonema Hua

摘要 为探明多花黄精的最佳组织培养体系,实现其快速繁殖,以多花黄精根状茎为外植体,研究不同处理的消毒效果及不同植物生长调节剂对多花黄精不定芽诱导、增殖和生根的影响。结果表明:70%乙醇溶液浸泡30s后再用0.1%HgCl2溶液浸泡30min可达最佳的消毒效果,污染率仅26%;细胞分裂素对于不定芽的诱导起主要作用,其中ZT的效果最优,KT最差,以培养基1/2 MS+ZT 1.0 mg/L+NAA0.2mg/L诱导不定芽的效果最佳,诱导率达87.9%,不定芽数为2.99个。细胞分裂素对不定芽的增殖有促进作用,其中6-BA的效果好于KT,以培养基1/2MS+6-BA 4.0mg/L+IAA 1.0mg/L的增殖效果最佳,增殖率为4.74倍。培养基1/4 MS+NAA 0.5mg/L适合诱导不定芽生根,生根率达88.75%,且生根良好。 The effects of different disinfection treatments and plant growth regulator combinations on induction, propagation and rooting of P. cyrtonerna Hua adventitious buds by taking rhizomes of P. cyrtonema Hua as explants to discuss the optimum tissue culture system of P. cyrtonema Hua and realize its rapid propagation. Results:The contamination rate of P. cyrtonema Hua explants treated with 70% ethanol solution for 30 s +0. 1% HgC12 solution for 30 min is 26G only. Cytokinin has the main effect on induction of adventitious buds and the best and poorest induction effect of tested cytokinins is ZT and KT respectively. The adventitious bud induction rate and number of explants cultured on the optimum medium with 1/2 MS+1.0 mg/L ZT+0.2 mg/L NAA can reach 87.9% and 2.99 respectively. Cytokinin has the promoting effect on propagation of adventitious buds and the promoting effect of 6-BA is better than KT. The propagation times of adventitious buds cultured on the optimum medium with 1/2 MS+ 4.0 mg/L 6-BA+I. 0 mg/L NAA are up to 4.74. The rooting rate of adventitious buds cultured on the optimum medium with 1/4 MS+0.5 mg/L NAA can reach 88.75%.
