摘要
为了解热带地区牛冠状病毒(Bovine coronavirus,BCoV)感染情况,对2019年1月~12月云南景洪采集的214份牛食道-咽喉部分泌物(esophagus-pharyngeal secretion,OP液)进行筛查,针对BCoV S蛋白基因片段进行RT-PCR扩增和测序,阳性样品分段扩增病毒全基因序列,进行同源性及遗传进化分析。结果显示,1份牛OP液样品中扩增到BCoV S蛋白基因目的片段622bp,且与中国、越南、美国、瑞典等国家分离的BCoV位于同一分支,属于冠状病毒β属A亚群病毒;全基因组序列分析该毒株与美国毒株US/OH3/2006相似性达到99.33%,二者具有较高的同源性;结构蛋白HE、S、M、N与US/OH3/2006毒株相似性分别为99.29%、99.36%、99.42%和100%,结构蛋白基因主要变异位点在HE基因,其次为S基因。研究结果为进一步开展BCoV分子流行病学以及检测研究提供科学依据。
In order to understand the infection of bovine coronavirus(BCoV)in tropical areas,214 bovine oesophageal pharyngeal secretions(OP fluid)collected in Jinghong,Yunnan province from January to December 2019 were screened.Reverse transcription-polymerase chain reaction(RT-PCR)was used to amplify and sequence the gene fragment of the BCoV S protein,the whole genome sequence was amplified in positive samples,and homology and genetic evolution were analyzed.The results showed that 622bp of BCoV S protein gene was amplified in one OP fluid sample,the gene was located in the same branch as BCoV isolated from China,Vietnam,the United States,Sweden and other countries.So the virus belongs to subgroup A virus of coronavirusβ.The whole genome sequence analysis showed that the similarity between the strain and the American strain US/OH3/2006 was 99.33%.The similarities of structural proteins HE,S,M and N with US/OH3/2006 strain were 99.29%,99.36%,99.42%and 100%respectively.The main variation site of structural protein gene was HE gene,followed by S gene.The results provide a scientific basis for further research on BCoV molecular epidemiology and detection.
