摘要
为了解吉林省延边地区边境县域游离蜱和牛携带东方泰勒虫情况,2019年4—8月在吉林珲春、图们、龙井及和龙市采集游离蜱和黄牛的静脉血,采用PCR方法检测东方泰勒虫主要表面蛋白MPSP基因特异性片段,确定东方泰勒虫感染情况,并通过基因测序、比对蜱源和牛源东方泰勒虫基因序列,分析其相关性。结果显示,采集的1183只游离蜱中,东方泰勒虫最低感染率为1.52%;采集的30份牛血液样本中,东方泰勒虫阳性率为33.33%。经基因序列分析,蜱源和牛源东方泰勒虫MPSP基因序列与NCBI GenBank中记录的东方泰勒虫基因序列同源性均达到了99.50%以上;经系统进化分析,蜱源和牛源东方泰勒虫基因序列在同一个分枝上,两个序列进化关系较近。结果表明,吉林延边边境地区牛感染东方泰勒虫可能来源于蜱类传播。
To investigate the situation of Theileria orientalisi carried by free ticks and cattle in border area of Yanbian area of Jilin Province,free ticks and bovine venous blood were collected in Hunchun,Tumen,Longjing and Helong counties of Yanbian Prefecture of Jilin province from Apr.to Aug.2019.The specific fragments of MPSP(major piroplasm surface proteins)gene of T.orientalisi were detected by PCR.The gene sequences of T.orientalisi from ticks and cattle were compared and their correlation was analyzed.As results,the minimum infection rate per 100 ticks(MIR)of T.orientalisi in collected 1183 free ticks was 1.52%.A total of 30 bovine blood samples were collected,and the positive rate of T.orientalisi was 33.33%.By gene sequence analysis,the homology of MPSP gene sequence of T.orientalisi originated from tick and cattle with that of T.orientalisi deposited in NCBI GenBank reached above 99.50%.Phylogenetic analysis showed that the gene sequences of T.orientalisi originated from tick and cattle in Yanbian area were clustered in the same branch of phylogeny tree,and these sequences were closely related.The results show that T.orientalisi infected in cattle in Yanbian area of Jilin Province may be caused by tick bites.
