摘要
目的对盐炙车前子多糖进行初步化学研究,并考察其对痛风性肾病大鼠的预防作用。方法采用水提醇沉法提取盐炙车前子多糖,苯酚-硫酸法、硫酸-咔唑法和考马斯亮蓝法测定盐炙车前子多糖的总糖含量、糖醛酸含量和蛋白质含量,PMP柱前衍生化法对多糖进行单糖组成分析,腺嘌呤和酵母联用制备痛风性肾病模型大鼠,研究盐炙车前子多糖的抗痛风性肾病作用。结果盐炙车前子多糖得率为10.90%,总糖含量为32.27%、糖醛酸含量为30.71%、蛋白质含量为3.46%,其主要由葡萄糖(Glc)、阿拉伯糖(Ara)和木糖(Xyl)组成,其摩尔比为甘露糖(Man)∶鼠李糖(Rha)∶半乳糖(Gal)∶Glc∶Ara∶Xyl∶岩藻糖(Fuc)=1.07∶1.49∶5.46∶14.94∶14.34∶42.86∶3.63,是一种阿拉伯木聚糖。盐炙车前子多糖可缓解痛风性肾病模型大鼠的肾组织病变,减少尿酸盐结晶沉积,降低肾质量指数,降低大鼠24 h尿蛋白、血尿酸(UA)、尿素氮(BUN)和肌酐(CRE)的水平。结论盐炙车前子多糖可改善肾功能,对痛风性肾病大鼠具有保护作用。
Objective To study the polysaccharides of Plantain seeds roasted with salt and its therapeutic effect on gouty nephropathy rats.Methods The polysaccharides were extracted by water extraction and alcohol precipitation.The total sugar content,uronic acid content and protein content of the polysaccharides were determined by phenol sulfuric acid method,sulfuric acid carbazole method and Coomassie brilliant blue method,respectively.The monosaccharide composition of polysaccharide was analyzed by PMP pre column derivatization method.The gouty nephropathy model rats were prepared by adenine and yeast.The anti-gouty nephropathy effect of polysaccharides from Plantain seeds was studied.Results The yield of polysaccharides from Plantain seeds was 10.90%,the total sugar content was 32.27%,the uronic acid content was 30.71%,and the protein content was 3.46%.It was mainly composed of glucose(Glc),arabinose(Ara)and xylose(Xyl).Its molar ratio was Man∶Rha∶Gal∶Glc∶Ara∶Xyl∶Fuc=1.07∶1.49∶5.46∶14.94∶14.34∶42.86∶3.63.It was an arabinoxylan.Salt-processed Plantain seeds polysaccharides alleviated the pathological changes of renal tissue in gouty nephropathy rats,reduced urate crystal deposition,decreased renal mass index,and lowered the levels of 24-hour urinary protein,blood uric acid(UA),urea nitrogen(BUN)and creatinine(CRE)in rats.Conclusion Salt-processed Plantain seeds polysaccharides can improve renal function and protect rats with gouty nephropathy.
