摘要
长根菇含有丰富的多糖、氨基酸、多酚和三萜类等成分,具有较高的食用价值和药用价值,是我国传统药用真菌之一。论文首先对超声辅助提取长根菇子实体多糖的工艺条件进行了优化;然后通过红外光谱初步考察了多糖的结构,最后考察了长根菇多糖的体外抗氧化活性。研究结果表明:长根菇子实体多糖的最佳提取工艺为超声提取时间41 min、料液比1:18、提取温度77℃,在该提取条件下,长根菇多糖的提取率达到10.33%;体外抗氧化研究表明长根菇多糖具有较好的体外抗氧化活性,但其抗氧化能力均低于Vc,多糖对DPPH·自由基清除能力、OH·自由基清除能力和三价铁离子还原能力的IC_(50)分别为4.32、0.79 mg/mL和4.40 mg/mL;红外光谱表明具有典型的多糖结构特征,其主要官能团有O—H、C—H、C=O、C—O—C和C—O—H等,且含有吡喃糖。研究结果为长根菇多糖产品的开发提供了数据和技术支撑,对相关功能性食品的开发提供了一定的参考价值。
Oudemansiella radicata is one of the traditional medicinal fungi in China,which is rich in polysaccharides,amino acids,polyphenols and triterpenoids.In this paper,the ultrasonic assisted extraction of polysaccharides from Oudemansiella radicata was optimized.The structure of the polysaccharide was investigated by infrared spectroscopy.The in vitro antioxidant activity of the polysaccharides was investigated.The results showed that the optimal extraction conditions were as follows:ultrasonic extraction time was 41 min,solid-liquid ratio was 1:18,extraction temperature was 77℃,under these conditions,the extraction rate of polysaccharide reached 10.33%.The results showed that the polysaccharides had good antioxidant activity in vitro,but its antioxidant capacity was lower than that of Vc.The IC_(50) values of DPPH·free radical scavenging ability,OH·free radical scavenging ability and reducing power ability of polysaccharide were 4.32 mg/mL,0.79 mg/mL and 4.40 mg/mL,respectively.The infrared spectrum showed that the polysaccharides had typical structure characteristics,including the main functional groups O–H,C–H,C=O,C–O–C and C–O–H,and containing pyranose.The research provides data and technical support for the development of polysaccharides from Oudemansiella radicata,and provides certain reference value for the development of related functional foods.
