摘要
目的 探讨miRNA-34a、miRNA-145以及miRNA-335在人脑胶质瘤组织中的表达及临床意义。方法 收集经病理证实的脑胶质瘤患者的脑脊液标本63例为病例组,同期非颅内肿瘤患者脑脊液标本20例为对照组。采用PCR检测miRNA-34a、miRNA-145以及miRNA-335在纳入群体脑脊液中的表达;采用Spearman秩相关分析miRNA-34a、miRNA-145以及miRNA-335的表达与在不同级别胶质瘤中的相关性。结果 病例组患者脑脊液中miRNA-34a、miRNA-145以及miRNA-335的表达水平显著低于对照组(P<0.05)。高级别胶质瘤患者脑脊液中miRNA-34a、miRNA-145以及miRNA-335的表达水平显著低于低级别胶质瘤患者(P<0.05);Spearman秩相关分析显示miRNA-34a、miRNA-145以及miRNA-335的表达与肿瘤级别呈显著负相关性(P<0.001)。结论 人脑脊液中miRNA-34a、miRNA-145以及miRNA-335的表达,可能可以作为诊断脑胶质瘤及术前判断病理分级的辅助方法。
Objective To investigate the expressions and clinical value of miRNA-34 a,miRNA-145 and miRNA-335 in human glioma.Methods The cerebrospinal fluid samples of 63 patients with pathologically confirmed gliomas were collected as the case group,and 20 patients with non intracranial tumors were collected as the control group.PCR was used to detect the expressions of miRNA-34 a,miRNA-145 and miRNA-335 in cerebrospinal fluid of human glioma patients and normal patients;Spearman rank correlation analysis was used to analyze the correlation between expressions of miRNA-34 a,miRNA-145 and miRNA-335 and different grades of glioma.Results The expression levels of miRNA-34 a,miRNA-145 and miRNA-335 in cerebrospinal fluid of patients in the case group were significantly lower than those in the control group(P<0.05).The expressions of miRNA-34 a,miRNA-145 and miRNA-335 in cerebrospinal fluid of patients with high-grade glioma were significantly lower than those in the low-grade glioma patients,(P<0.05).Spearman rank correlation analysis revealed that there was a significant negative correlation between the expressions of miRNA-34 a,miRNA-145 and miRNA-335 and tumor grade(P<0.001).Conclusion The expressions of miRNA-34 a,miRNA-145 and miRNA-335 of human cerebrospinal fluid might be assisted methods to diagnose human glioma and judge the pathological grade.
