摘要
目的:探究加味柴芍六君汤逆转慢性萎缩性胃炎(CAG)大鼠胃黏膜病理改变的炎症相关生物学机制。方法:将20只SD雄性大鼠随机分为正常组6只,造模组14只。采用化学诱导+饥饱失常的方法建立CAG大鼠模型,共16周,造模结束后随机取2只造模组大鼠进行模型评价。成模大鼠随机分为模型组和加味柴芍六君汤组,每组6只。正常组和模型组给予等体积(10mL/kg)生理盐水灌胃,加味柴芍六君汤组给予0.69g/mL加味柴芍六君汤灌胃,干预4周。观察胃黏膜病理形态变化,检测3组大鼠血清炎症因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的含量及胃黏膜组织HIF-1αmR-NA表达水平。结果:正常组胃固有层腺体排列整齐,未见腺体萎缩、减少;模型组大鼠胃黏膜固有层腺体排列紊乱,局部腺体萎缩、减少,腺腔增大,炎性细胞浸润,可见多处散在出血点;加味柴芍六君汤组腺体排列较整齐,腺体萎缩改善,仍可见散在出血点。模型组大鼠血清IL-1β、IL-6含量显著升高(P<0.01),胃黏膜组织HIF-1αmRNA表达水平较正常组上升(P<0.05)。加味柴芍六君汤组大鼠血清IL-1β、IL-6含量显著降低(P<0.01),HIF-1αmRNA表达水平较模型组显示出下降的变化趋势(P>0.05)。结论:加味柴芍六君汤改善CAG胃黏膜组织病理的机制可能是通过降低血清IL-1β、IL-6含量,下调胃黏膜缺氧诱导因子-1α(HIF-1α)过表达,从而改善胃黏膜局部缺氧环境,减轻炎症反应。
Objective:To explore the inflammation related biological mechanism of modified Chaishao Liujun Decoction in reversing the pathological changes of gastric mucosa in rats with chronic atrophic gastritis(CAG).Methods:20 male SD rats were randomly divided into the normal group(n=6)and the modeling group(n=14).The CAG rat model was established by chemical induction plus abnormal hunger and satiety for 16 weeks.After the modeling,two model rats were randomly selected for model evaluation.The model rats were randomly divided into the model group and the study group,with 6 rats in each group.The normal group and the model group were given equal volume of normal saline(10 mL/kg)by gavage,and the study group was treated with modified Chaishao Liujun Decoction(0.69 g/mL)by gavage for 4 weeks.The pathological changes of gastric mucosa were observed,the contents of serum inflammatory factors of interleukin-1β(IL-1β)and interleukin-6(IL-6),as well as the mRNA expression level of inducible factor-1α(HIF-1α)were detected in the three groups.Results:In the normal group,the glands of gastric lamina propria were arranged orderly,and there was no atrophy or reduction of glands;in the model group,the glands in the lamina propria of gastric mucosa were arranged disorderly,local glands atrophied and decreased,the gland cavity increased,inflammatory cells infiltrated,and multiple scattered bleeding points could be seen;in the study group,the glands were arranged neatly,the gland atrophy was improved,and scattered bleeding points could still be seen.The serum contents of IL-1βand IL-6 were significantly increased(P<0.01),and the mRNA expression level of HIF-1αwas also increased in the model group than those in the normal group(P<0.05).The serum contents of IL-1βand IL-6 were significantly decreased(P<0.01),and the mRNA expression level of HIF-1αshowed decreasing tendency in the study group compared with those in the model group(P>0.05).Conclusion:The mechanism of modified Chaishao Liujun Decoction in improving the histopathology of CAG gastric mucosa may be related to reducing serum contents of IL-1βand IL-6,down-regulating gastric mucosal HIF-1αexpression,thus to improve the local hypoxic environment of gastric mucosa and reduce inflammatory response.
