四妙勇安汤“异病同治”冠状动脉粥样硬化性心脏病和血栓闭塞性脉管炎的机制研究 被引量:4

摘要 目的运用网络药理学方法,研究四妙勇安汤在“异病同治”理论指导下治疗冠状动脉粥样硬化性心脏病和血栓闭塞性脉管炎的共同机制。方法通过TCMSP数据库检索四妙勇安汤中4味中药相关的所有单体和作用靶点,运用UniProt数据库进行标准化,并用Cytoscape 3.7.2软件构建“中药-化合物-靶点”作用网络;通过GeneCards和NCBI数据库获取冠状动脉粥样硬化性心脏病和血栓闭塞性脉管炎的相关基因,并与药物作用靶点取交集,利用Cytoscape 3.7.2软件构建三者的靶点关联网络;输入共有靶点于STRING数据库,构建PPI网络;进行GO(gene ontology)与KEGG通路富集分析(Kyoto encyclopedia of genes and genomes)。结果根据口服生物利用度为≥30%和类药性指数为≥0.18,筛选得四妙勇安汤活性化合物,计120个,并预测到205个潜在靶点,对两种疾病共有靶点共48个。GO功能富集生物过程条目计46项(P<0.01),同时在KEGG通路富集分析筛选上有147条信号通路(P<0.01),其中靠前的多条信号通路与免疫炎症密切相关。结论该研究表明四妙勇安汤主要是通过调控炎症相关信号通路达到“异病同治”冠状动脉粥样硬化性心脏病和血栓闭塞性脉管炎的主要机制,并通过荟萃分析证实其“异病同治”的疗效和炎症调节作用。 Objective To explore the mechanism of Simiaoyong’an Decoction in treating atherosclerotic heart disease and thromboangiitis obliterans through the theory of"same treatment to different diseases"by using network pharmacology Methods The TCMSP database was used to retrieve all of monomers and targets related to the four herbs in Simiaoyong’an Decoction.The database of UniProt was used for standardization.Meanwhile,Cytoscape 3.7.2 was used to build"traditional Chinese medicine-compound-target"action network;we used genecards and NCBI database to obtain the related genes of atherosclerotic heart disease and thromboangiitis obliterans,and took intersection with drug and targets,then used Cytoscape 3.7.2 to build the target association network of the them.PPI network with common target was built by the database of string.Gene Ontology and KEGG pathway enrichment analysis were carried out.Results According to the oral bioavailability≥30%and drug like index≥0.18,120 active compounds of Simiaoyong’an Decoction were screened,and 205 potential targets were predicted.There were 48 common targets for the two diseases from the decoction.There were 46 items of go functional enrichment in biological process(P<0.01).Meanwhile,147 signal pathways(P<0.01)were obtained through KEGG pathway enrichment analysis,and multiple signal paths in the front of which were closely related to inflammation.Conclusion This study showed the main mechanism of Simiaoyong’an Decoction in treating atherosclerotic heart disease and thromboangiitis obliterans by regulating inflammation related signaling pathways,and meta-analysis confirmed the efficacy of"same treatment to different diseases"and its role in inflammation regulation.
