摘要
为获得较高的槟榔多酚得率,需探明其最佳提取工艺参数。通过福林酚比色法测定槟榔多酚得率,采用单因素实验与响应面法结合优化槟榔多酚提取工艺参数。结果表明,3个提取工艺的因素参数对槟榔多酚得率影响由大到小依次为:提取温度、乙醇体积分数、提取时间。在固定料液比0.100 g·mL^(-1)的条件下,经软件分析预测得出多酚最佳提取工艺条件为:提取温度60℃、乙醇体积分数64%、提取时间70 min。在此工艺条件下,槟榔多酚得率为(20.55±0.706)mg·g^(-1),与回归方程模型的理论预测值20.98 mg·g^(-1)相近。优化后的提取工艺可稳定提高多酚得率,利于进行槟榔多酚的相关研究。
In order to obtain high yield of polyphenols from betel nut,it is necessary to investigate the optimum extraction technology parameters.The yield of polyphenols was determined by Folin-Ciocalteu method,and the parameters of extraction technology of polyphenols from betel nut were optimized by combining single factor experiment with response surface method.The results showed that the effects of various factors on the yield of polyphenols from betel nut were,in descending order:extraction temperature,ethanol volume fraction and extraction time.Under the condition of fixed solid-liquid ratio of 0.100 g·mL^(-1),the optimal extraction technology of betel nut’s polyphenols predicted by software analysis was as follows:extraction temperature 60℃,ethanol volume fraction 64%and extraction time 70 min.Under the optimal conditions,the yield of polyphenols was(20.55±0.706)mg·g^(-1),which was close to the theoretical predicted value of 20.98 mg·g^(-1).The optimized extraction technology,which improves stably the yield of polyphenols,is conducive to the related research of betel nut’s polyphenols.
