融水香菇多糖结构表征及体外抗氧化、降糖活性 被引量:24

Structural characterization,in vitro anti-oxidative effect and hypoglycemic activity of lentinan from Rongshui county

摘要 以产自广西融水的香菇为原料,研究其香菇多糖(Lentinan,LNT)的结构、体外抗氧化及降糖活性。采用傅里叶红外光谱、核磁共振、扫描电镜及刚果红实验对LNT进行结构分析。通过测定羟基自由基(·OH)、1,1-二苯基-2-三硝基苯肼(DPPH·)清除率及总还原力评价LNT抗氧化活性。采用测定α-葡萄糖苷酶的抑制活性及胰岛素抵抗HepG2(IR-HepG2)细胞葡萄糖消耗量研究LNT的降糖活性。结果表明LNT由β-吡喃糖构成,具有三股螺旋结构,其抗氧化活性与多糖浓度呈正相关,当LNT浓度为8mg/mL时,·OH与DPPH·基清除率分别为99.35%、90.30%,总还原力吸光度值为0.99;LNT对α-葡萄糖苷酶的活性有抑制作用,在0~8mg/mL浓度范围内,其抑制率最大为75.09%;LNT能提高IR-HepG2细胞的葡萄糖消耗量,较模型组显(7.99mmol/L)著升高了91.50%(^(**)p<0.01),提升至15.30mmol/L。本研究表明LNT具有较好的抗氧化能力,能够抑制α-葡萄糖苷酶的活性,减缓葡萄糖的吸收,增加葡萄糖消耗、减弱胰岛素抵抗,具有降低血糖的作用,为LNT开发利用提供了一定的理论与数据支撑。 The structure,antioxidant activity and hypoglycemic activity of lentinan(the origin of Lentinus edodes :Rongshui County,Guangxi)were investigated. The structure of lentinan(LNT)was identified by Fourier infrared technology(FT-IR),nuclear magnetic resonance technology(NMR),scanning electron microscope technology(SEM)and Congo red experiment. The antioxidant activity of LNT was evaluated by measuring the scavenging rates of hydroxyl radical( · OH),2,2-diphenyl-1-methylhydrazine radical(DPPH ·)and total reducing power. At the same time,by analyzing the inhibitory activity of LNT on α-glucosidase and its effect on glucose consumption in insulin resistance of HepG2 cells,the hypoglycemic activity of LNT was studied. The results showed that the LNT was β-pyranose with a triple helix structure. LNT had antioxidant activity which was positively correlated with polysaccharide concentration. When the concentration of LNT was 8mg/mL,the scavenging rates of · OH and DPPH · were 99.35% and 90.30% respectively,and the absorbance value of total reducing power was 0.99. LNT could inhibit the activity of α-glucosidase,the maximum inhibition rate of LNT was 75.09% in the concentration range of 0 ~ 8mg/mL. LNT could increase the glucose consumption of insulin resistant HepG2 cells by 91.50% to 15.30mmol/L compared with the model group(7.99mmol/L,^(**)p < 0.01). It indicated that LNT had good antioxidant capacity and could inhibit α-glucosidase activity,slow down the absorption of glucose,increase glucose consumption,weaken insulin resistance,and thus had the effect of lowering blood sugar. A certain theoretical and data support was provided for the development and utilization of LNT.
