纯钛表面制备纳米氧化镁薄膜的抗菌性能研究 被引量:2

摘要 目的:在纯钛表面制备不同厚度纳米氧化镁薄膜,对体外抗菌效果进行评价。方法:通过控制磁控溅射的时长,制备100 nm、200 nm以及300 nm三种不同厚度的纳米氧化镁薄膜。采用菌落计数,细菌形态观察方法,探究纯钛表面的不同厚度纳米氧化镁薄膜对牙龈卟啉单胞菌生长的影响。同时,通过CCK-8方法检测纳米氧化镁薄膜对MC3T3-E1细胞的细胞毒性。结果:纳米氧化镁薄膜具备较好抑菌性能,随薄膜厚度增加其抗菌效果逐渐增加。MC3T3-E1细胞在不同厚度纳米氧化镁薄膜组和空白组样本表面的存活率无差异,表明纳米氧化镁薄膜无细胞毒性。结论:纳米氧化镁薄膜能有效抑制牙龈卟啉单胞菌的生长,抵制生物膜的形成,为预防种植体周围炎提供新的思路。 Objective:To prepare the nanometer magnesium oxide films with different thickness on pure titanium and to evaluate the antibacterial effect in vitro.Methods:By controlling the time of magnetron sputtering,three kinds of nanometer magnesium oxide films with different thickness of 100 nm,200 nm and 300 nm were prepared.The antibacterial effect of nanometer magnesium oxide film with different thickness on porphyromonas gingivalis was investigated by colony counting and bacterial morphology observation.The MC3T3-E1 cells were inoculated onto the surface of each sample and cultured for 24 hours,48 hours and 72 hours.Cell activity of each group was detected by CCK-8.Results:The antibacterial rate of 100 nm nano magnesium oxide film was 78.14%,the antibacterial rate of 200nm and 300nm nanomagnesium oxide films was 87.16%and 99.86%respectively.The nanometer magnesium oxide film has excellent antibacterial properties,and the antibacterial effect gradually increases with the increase of the film thickness.Nanometer magnesium oxide film with a thickness of 300nm could nearly completely inhibit the growth of Porphyromonas gingivalis.The survival rate of MC3T3-E1 cells on the surface of the nano-magnesium oxide films showed no significant differences compared with the blank group,indicating that the nano-magnesium oxide films has no cytotoxicity.Conclusion:The nano-magnesium oxide film can effectively inhibit the growth of Porphyromonas gingivalis and resist the formation of biofilm,which provides new ideas for preventing peri-implant inflammation.
