摘要 目的:通过测定高剂量地塞米松对犬血常规、血液流变学指标的影响,反映高剂量地塞米松对血液成分及性质的影响,评价应用高剂量地塞米松的注意事项。方法:试验持续7 d,选取中华田园犬8只,每天08:30对实验犬进行颈静脉采血,第1~4天09:00肌注地塞米松(2 mg/kg·体重),第5~7天仅采血,第1~7天所采样本依次作为肌注前、肌注地塞米松1~4 d及恢复1~2 d时的样本。结果:与肌注前相比,各肌注地塞米松时间点(1~4 d)及恢复1、2 d时白细胞计数、粒细胞计数均显著升高(P<0.05),肌注地塞米松2~4 d时淋巴细胞计数显著降低,肌注地塞米松4 d及恢复1~2 d时红细胞计数、血红蛋白浓度及红细胞压积显著降低(P<0.05),各肌注地塞米松时间点(1~4 d)与恢复1、2 d时全血黏度低、中、高切值显著降低(P<0.05)。结论:高剂量地塞米松使犬血液中红细胞含量、淋巴细胞含量、全血黏度降低,中性粒细胞含量升高。表明高剂量地塞米松影响机体血液系统代谢,在临床上应用高剂量地塞米松时应严格把控用药剂量及时间,尽可能地减小负面作用。 Objective: By measuring the effects of high dose dexamethasone on blood routine and hemorheology indexes in dogs, the effects of high dose dexamethasone on blood components and properties were reflected, and the precautions for application of high dose dexamethasone were evaluated. Methods: The experiment lasted for 7 days. Eight Chinese pastoral dogs were selected and blood was collected from jugular vein at 08:30 every day. Dexamethasone(2 mg/kg, body weight) was intramuscularly injected at 09:00 on the 1 st, 2 nd, 3 rd and 4 th day, and only blood was collected on the 5 th, 6 th and 7 th day. The samples collected from the 1 st day to the 7 th day were used as samples before intramuscular injection, 1-4 d after intramuscular injection, and 1-2 d after recovery. Results: Compared with former intramuscular injection, white blood cell count and granulocyte count were significantly increased at each intramuscular injection time point(1-4 d) and 1-2 d after recovery(P<0.05). Lymphocyte count was significantly decreased at 2-4 d after intramuscular injection of dexamethasone. Erythrocyte count, hemoglobin concentration and hematocrit were significantly decreased at 4 d after intramuscular injection of dexamethasone and 1,2 d after recovery(P<0.05). Whole blood viscosity at each intramuscular injection time point(1-4 d) and 1, 2 d after recovery were significantly decreased(P<0.05). Conclusion: High-dose dexamethasone reduced the red blood cell content, lymphocyte content, whole blood viscosity, and increased neutrophil content. It is indicated that high dose dexamethasone affects the metabolism of blood system. In clinical application of high dose dexamethasone, the dosage and time should be strictly controlled to minimize the negative effect.
