摘要
目的建立一种固相萃取-高效液相色谱联合检测银线莲中核苷酸的分析方法。方法采用固相萃取柱对银线莲中的核苷酸成分进行净化、富集,用高效液相色谱法测定含量。色谱柱为:Agilent TC-C_(18)(4.6 mm×250 mm,5μm);流动相:A-0.008%甲酸/10 mmol·L^(-1)甲酸铵,B-甲醇,梯度洗脱;流速:0.80 mL·min^(-1),检测波长:260 nm。结果在最佳的萃取条件下,测得银线莲中核苷酸的含量UMP、GMP和AMP分别为277.8μg·g^(-1)、35.0μg·g^(-1)和147.4μg·g^(-1),加样回收率为88.4%~108.7%。结论该方法具有操作简便、灵敏准确和高效环保等优点,并成功应用于银线莲中3种核苷酸物质的萃取和测定。
OBJECTIVE To establish a method for determination of nucleotides in Goodyera hachijoensis Yatabe by solid-phase extraction combined with high-performance liquid chromatography(HPLC).METHODS According to the literature,the content of nucleotides in Goodyera hachijoensis Yatabe was determined by HPLC after purification and enrichment by solid phase extraction column.The chromatographic column was Agilent TC-C_(18)(4.6 mm 4×250 mm,5μm).The mobile phase was A-0.008%formic acid/10 mmol·L^(-1) ammonium formate,B-methanol,performed as gradient elution.The flow rate was 0.80 mL·min^(-1) and detection wavelength was 260 nm.RESULTS Under the aforementioned extraction conditions,the conetnts of UMP,GMP and AMP were 277.8μg·g^(-1),35.0μg·g^(-1) and 147.4μg·g^(-1) respectively,with an average recovery from 88.5%to 109.7%.CONCLUSION The proposed method showed the advantages of simplicity,efficiency,good recovery and environmental protection.The method was also successfully applied to nucleotides extraction and determination,which was firstly discovered in Goodyera hachijoensis Yatabe.
